摘 要:目的:为了进一步研究蝉拟青霉总多糖和腺苷单用或分别联合顺铂(DDP)或阿霉素(ADM)在体外的抗肿瘤作用。方法:用MTT法检测DDP、ADM、总多糖、总腺苷单独及联合用药对人肺腺癌(PAA-1)细胞的生长抑制能力;用流式细胞仪检测其对体外培养细胞系细胞周期的影响;用光镜检查其对体外培养PAA-1细胞系集落形成率的影响;用流式细胞仪检测其对体外培养淋巴细胞亚群分化的影响。结果:DDP、ADM、总多糖、腺苷对人肺腺癌(PAA-1)细胞生长抑制的IC50分别为9.05μg/mL、36.21μg/mL、10.86mg/mL、7.81mg/mL。DDP或ADM和多糖联用对PAA-1细胞的生长抑制主要表现出拮抗效应;0.5mg/mL、0.25mg/mL腺苷与4μg/mL阿霉素联合作用时,显示出拮抗效应,而当8mg/mL腺苷与4μg/mL阿霉素联用时,显示协同效应;不同浓度腺苷单用与和DDP联合使用相比,对细胞生长影响无显著性差异;腺苷和ADM联合作用,随着腺苷浓度的增加,G0/G1期细胞逐渐减少,而G2/M期相对增加。对体外培养PAA-1细胞系集落形成率多糖组和阴性对照组未见明显差异,不同浓度腺苷组的集落形成率略低于阴性对照组,随着腺苷浓度逐渐增加,集落形成率逐渐下降。多糖对体外培养淋巴细胞亚群的分化无影响。结论:总多糖、腺苷具有一定的体外抗肿瘤作用,总多糖与DDP或ADM联用不能增加其体外的抗肿瘤作用,腺苷与ADM联用可增加其体外的抗肿瘤作用。Objective: To further study anti -tumor effect in vitro of polysacchafides and adenosine respectively, or combined with eisplatin (DDP) or adriamycin (ADM). Methods : Detection of inhibition of human lung adenocareinoma ( PAA - 1 ) eell growth with DDP, ADM, polysaechafides and adenosine respectively or combined administration by MTT assay, eel] cycle by flow eytometry on cultured cell lines, colony - forming rate of PAA - 1 cell in vitro by light micro- scope examination, and lymphocyte subsets by flow eytometry. Results : IC50 of DDP, ADM, total polysaccharides, and adenosine on growth inhibition of human lung adenocarcinoma ( PAA - 1 ) cell were 9. 05lxg/mL, 36.21p,g/mL, 10. 86mg/mL and 7.81mg/mL. It is antagonist of Polysaccharide combined with ADM or DDP on growth inhibition of PAA - 1 cells. Adenosine with 8mg/mL combined with ADM with 4txg/mL indicate synergism. It is no different between different concentrations of adenosine alone and combined with DDP on cell growth. With the increase of concentration of a- denosine, G0/G1 phase cells decreased, while the G2 / M phase relative increase when adenosine combined and ADM. It is no different between polysaccharide group and negative control group on colony - forming rate of PAA - 1 eel] in vitro. Adenosine groups were slightly lower than negative control group on colony -forming rate. With a gradual increase in the concentration of adenosine, the rate of colony - forming gradually decreased. Polysaecharides had no effect on on lymphocyte subsets of PAA - 1 cell in vitro. Conclusion: The polysaccharides and adenosine has a certain anti - tumor effects in vitro. Polysaceharide combined with ADM or DDP can not increase their anti -tumor effects in vitro, adenosine combined with ADM can increase anti - tumor effects in vitro.
