摘 要:目的研究硒化壳聚糖对体外培养的NB4细胞增殖和凋亡的影响,探讨这种作用与端粒酶活性和survivin蛋白的关系。方法MTT法检测细胞增殖;细胞形态学观察和流式细胞仪检测细胞凋亡;PCR-ELISA法检测端粒酶活性;western blot法检测survivin蛋白含量。结果50~200 mg/L硒化壳聚糖可时间剂量依赖性地抑制NB4细胞增殖,诱导其凋亡,降低端粒酶活性和下调survivin蛋白含量。结论硒化壳聚糖可通过抑制端粒酶活性,下调survivin蛋白含量,诱导细胞凋亡从而抑制体外培养的NB4细胞增殖。Objective To investigate the effect of selenium chitosan on proliferation and apoptosis in NB4 cells and explore its relationship with the telomerase activity and survivin protein.Methods MTT was used to assess the cell proliferation;Microscopic observation and flow cytometry were used to assess the induced apoptosis;Polymerase chain reaction enzyme-linked immunoassay(PCR-ELISA) was used to measure the telomerase activity;Western blot was used to examine the expression level of survivin protein.Results Exposure to 50~200 mg/L of selenium chitosan for 24~72 h inhibited proliferation and induced apoptosis in NB4 cells in dose and time dependent manners.The telomerase activity and expression of survivin protein were strongly inhibited in selenium chitosan treated NB4 cells.Conclusion Selenium chitosan inhibites NB4 cells proliferation in vitro via reducing telomerase activity and down-regulating survin protein and induces apoptosis.
