山西汉族人群Y-STR基因座DYS714的多态性研究

摘　　要：目的研究山西汉族群体Y—STR基因座DYS714的多态性，并探讨其在法医学中的应用价值。方法应用PCR扩增方法结合DNA分析序列技术对120例山西地区汉族男性个体DYS714基因座进行分析。结果DYS714基因座在山西汉族男性个体中观察到5个等位基因，核心序列为ttttc，重复14—18次，基因频率分别为0．15、0．1583、0．2167、0．3333、0．1416，基因多样性为0．7808。个人识别能力（DP）和非父排除率（PE）均为0．7808。28组家系调查无突变。结论DYST14基因座具有很好的多态性和稳定性。Objective To investigate a Y-STR locus （DYS？14） on male Y chromosome to obtain the allele frequencies of DYS？14 in Han population, Shanxi Province and to evaluate its application in forensic casework. Methods The candidates of Y-STR loci were selected from the GDB database. Primer pairs were designed on both side of the repeat region. The PCR products were analyzed with polyacrylamide gel electrophoresis. Each allele of this Y-STR loci was se quenced and the allele ladders were constructed. Alleles of these Y-STR loci were named according to recommendations of the International Society of Forensic Genetics. Results Our re- sults show that the DYS？14 locus consists of repeats of a core sequence （ttttc）. Five alleles were found among 120 males in Shanxi Province, and were corresponingly named 14,15,16, 17,18, the allele frequency being 0.15,0.1583,0. 2167,0. 3333,0.1416 respectively. Genetic diversity （h） of DYST14 was 0. 7808. The power of discrimination （DP） and the power of exclusion （PE） were both 0. 7808. No amplified products could be detected from 20 female samples. Mutations were not found from 28 father-offspring pairs. Conclusion DYST14 have a high level of genetic Polymorphism.
