摘　　要：目的探讨生物素化抗AIB1单抗在肿瘤细胞AIB1检测中的应用。方法用生物素N-羟基丁二酰亚胺酯(BNHS)对纯化抗体1A2E1进行标记,获得了Biotin-1A2E1,通过免疫荧光化学法应用该生物素化单克隆抗体检测肿瘤细胞中AIB1蛋白的表达情况。结果应用所制备的生物素化抗体检测靶细胞中AIB1蛋白,免疫荧光结果显示敏感性高,特异性强。结论可成功应用该抗体检测肿瘤细胞AIB1蛋白,为肿瘤的诊断、治疗提供新的研究思路。Objective Investigate application of biotinylatedanti-AIB1 monoclonal antibody for AIB1 detection in tumour cells.Methods We use the BNHS reagents to mark the McAb 1A2E1 and harvest Biotin-1A2E1 with its valence-1∶3200.We apply Biotin-1A2E1 to detect AIB1 protein in tumour cells by immunofluorescence chemical method.Results Utilizing the biotinylated McAb to detect AIB1 antigen of target cells,the results show that the McAb kept strongly specific and sensitivity.Conclusion It is successful to detect the target cells which express AIB1 antigen by the biotinylated antibody,and it may be of potential significance in the diagnosis and therapy for human cancer.
