摘  要:目的观察凉血活血复方水醇粗提液对人永生化表皮细胞株(HaCaT)和人脐静脉血管内皮细胞株(ECV304)端粒酶活性的影响,探讨该复方治疗银屑病可能的作用机制。方法将不同浓度的凉血活血复方水醇粗提液分别地作用于体外培养的HaCaT和ECV304细胞株,应用端粒重复序列扩增-酶联吸附(TRAP-ELAISA)试剂盒检测两株细胞的端粒酶活性。结果凉血活血复方分别作用两株细胞48h,细胞端粒酶活性在所测定实验浓度范围内,随浓度的增加对细胞的端粒酶活性的下调作用逐渐增强。各加药组与对照组在统计学上有显著性差异,且各加药组之间差异亦有统计学意义(P<0.01)。结论凉血活血复方以剂量依赖性抑制端粒酶活性。Objective To investegate the effects of the traditional Chinese medicine, the crude extract of Liangxue Huoxue complex prescription with water and alcohol on telomerase activity in keratinocyte HaCaT and human umbilical vein endothelial cell ECV304, and to explore the effective concentration and the mechanism of there antipsoriatic effects. Methods In present study, after simulating keratinocyte cell hyper-proliferation with human immortal epidermic cell, HaCaT cell line, and capillary vessel with human umbilical vein endothelial cell, ECV304 cells line, cultured HaCaT and ECV304 cells in vitro were exposed respectively to the crude extract of Liangxue Huoxue complex prescription at different concentrations. TRAPELISA method was used to detect the telomerase activity. Results Telomerase activity of HaCaT and ECV304 cells was downregulated at concentration-dependent manner. Telomerase activity of ECV304 cells was down-regulated at concentrations both sufficient(20mg/mL and 50mg/mL) and insufficient (10mg/mL) to induce apoptosis. Each experimental group showed statistical significance compared with control group, and significant difference between experimental groups was also displayed (P0.01). Conclusion Liangxue Huoxue complex prescription could inhibit telomerase activity in a dose-dependent manner.
