健脾复方对人大肠癌移植瘤裸小鼠血管内皮生长因子及肿瘤微血管密度的影响

摘　　要：目的研究健脾复方对人大肠癌移植瘤裸小鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)和肿瘤微血管密度(microvessel density,MVD)的影响。方法对60只BALB/C裸小鼠建立人大肠癌原位移植瘤模型,并按1:1:1:1:1随机分为健脾复方+5-Fu组、健脾复方组、塞来昔布组、5-Fu组和模型对照组,分别予相应的干预措施处理8周。分别观察VEGFmRNA(采用RT-PCR法)和MVD(免疫组化法)的表达,并对影响其表达的相关因素进行分析。结果与模型组比较,健脾复方+5-Fu、健脾复方、塞来昔布和5-Fu的治疗均可降低VEGFmRNA的表达(P<0.01),健脾复方+5-Fu、健脾复方可进一步降低MVD表达,且与5-Fu组和模型对照组的差异有显著性(P<0.05)。回归分析显示VEGFmRNA和MVD的表达与不同的治疗方案具有相关性。结论健脾复方单独使用或联合5-Fu,均可显著抑制大肠癌瘤块VEGFmRNA和MVD的表达,提示健脾复方有抑制肿瘤生长、复发、转移的潜能。Objective To investigate the effects of ＂Jianpi Complex Prescription＂ on expression of vascular endothelial growth factor （VEGF） and microvessel density （MVD） in nude mice of human colorectal cancer grafts. Methods Human colorectal cancer orthotopic transplanted tumor model was established in 60 BALB/C nude mice. The nude mice were randomized into Jianpi Complex Prescription ＋5-Fu group, Jianpi Complex Prescription group, celecoxib group, 5-Fu group and model control group and received given treatments for 8 weeks. VEGF mRNA （by RT-PCR assay） and MVD （by EnVision Immunohistochemisty assay） were measured. The influence factors of both parameters were analyzed. Results The expression of VEGF mRNA was reduced in Jianpi Complex Prescription ＋5-Fu group, Jianpi Complex Prescription group, eelecoxib group and 5-Fu group compared with model control group. MVD was decreased in Jianpi Complex Prescription ＋5-Fu group, Jianpi Complex Prescription group compared with 5-Fu group and model control group separately. The sole significant factor associated with expres- sion of VEGF mRNA and MVD was therapeutic regimen. Conclusion ＂Jianpi Complex Prescription＂ with or without 5-Fu can reduce the expression of VEGF mRNA and MVD, indicating that ＂Jianpi Complex Prescription＂ has potential to inhibit the growth, recurrence and metastasis of colonrectal cancer.
