摘  要:目的探讨美曲磷酯对血管性痴呆大鼠学习记忆的影响。方法采用两血管阻断法制作大鼠血管性痴呆模型,大鼠随机分为假手术组、血管性痴呆模型组、脑复康(100 mg.kg-1.d-1,ig×60)阳性药物对照组、美曲磷酯(7 mg.kg-1.d-1,ig×60)治疗组。Morris水迷宫检测大鼠学习记忆能力,定位航行实验以逃避潜伏期及游泳总路程反映其学习成绩,空间探索实验以大鼠穿越平台次数、有效区停留时间及游泳总路程反映其空间记忆成绩;显微镜观察大鼠海马脑区组织结构改变。结果模型组大鼠逃避潜伏期、游泳总路程均明显大于假手术组(P<0.05),有效区停留时间和穿越平台次数显著减少(P<0.05)。美曲磷酯能显著缩短血管性痴呆大鼠逃避潜伏期、游泳总路程(P<0.05),与脑复康效果相似而与假手术组处于同一水平;同时使血管性痴呆大鼠穿越平台的次数增加及有效区停留时间延长(P<0.05)而与假手术组接近,脑复康则对血管性痴呆大鼠穿越平台的次数及有效区停留时间无明显影响。病理学观察显示模型组大鼠海马各区细胞排列疏松、紊乱,细胞层及细胞数明显减少,细胞变性严重;美曲磷酯组则细胞排列紧密、均匀,细胞层及细胞数明显增多,细胞变性减轻与脑复康组形态变化相似。结论美曲磷酯可改善血管性痴呆大鼠的学习记忆能力,且改善空间记忆能力优于脑复康。Objective To study the effect of metrifonate on learning and memory of rats with vascular dementia. Methods The VD rat model was established by 2VO occlusion. The wistar rats were randomly divided into sham operation group, vascular dementia model group,the metrifonate treatment group(7 mg·kg^-1·d^-1,ig×60) and piraeetam group ( 100mg·kg^-1·d^-1,ig× 60). The ability of learning and memory was analyzed by Morris water maze test. The learning score was observed by stage of latency and pathway in the place navigation test, and the space memory was checked by the times that acrossed the target and the time stay in coverage area and pathway in the probe test. The hippocampal morphological changes were observed by microscope. Results Compared with sham operation group, stage of latency and pathway of vascular dementia model group increased(P 〈 0.05). The time of acrossing coverage area and the times acrossing the target were declined(P 〈 0.05 ). Stage of latency and pathway of rats with vascular dementia that treated by metrifonate was more shorter( P 〈 0.05 ) , similar with the sham - operation group and on the same level with piraeetam group, and the time of acrossing coverage area and the times aerossing the target were increased markedly(P 〈0.05) but showed no difference as compared to sham operation group; the time of acrossing coverage area and the times aerossing the target of rats with vascular dementia that treated by piracetam had little change. Pathological observation showed that the number of necrosis in hippoeampal area was reduced and the disorder in VD model group rangement compared with model group, metriforate could significantly improve the pathological changes of hippocampal neurons, whicth was similar with piracetam. Conclusion Metrifonate can improve the ability of learning and memory of the rats with vascular dementia, and improve the ability of space memory more remarkablly than piraeetam group does.
