Vasoactive substance and nitric oxide synthase expression changes of endothelium in arterioles of hindlimb muscle from tail suspended/recovered rats

摘  要:目的阐明模拟失重下血管内皮的变化。方法以尾部悬吊方法模拟失重影响,用免疫组织化学法观察对照(CON)、尾部悬吊4周(SUS-4)和恢复1周(REC-1)大鼠后肢肌肉小动脉内皮血管活性物质及一氧化氮合酶(NOS)活性的改变。结果在腓肠肌(抗重力肌),模拟失重4周后,NOSⅡ、NOSⅢ、P物质(SP)及内皮素-1(ET-1)免疫反应(Immunoreactive,IR)阳性的小动脉长度均较对照组显著减小。其中,又以ET-1-IR阳性小动脉长度的相对变化为最大,减少了60%(P<0.01),SP-IR阳性者减少了27%(P<0.01),NOSⅡ-IR与NOSⅢ-IR阳性者分别减少了11%(P<0.05)与46%(P<0.01)。解除模拟失重1周后,则呈现相反改变,SP-IR、ET-1-IR阳性者较对照组分别增加了77%及17%(P<0.01),而NOS则以Ⅱ型-IR的阳性者变化较大,增加33%(P<0.01),Ⅲ型-IR阳性者增加了28%(P<0.01)。在跖肌(非抗重力肌),各组免疫反应阳性小动脉的长度普遍低于腓肠肌;模拟失重4周后,SP、ET-1免疫反应阳性小动脉长度较对照组显著减小,亦以ET-1-IR阳性者?Objective To elucidate the effect of simulated weightlessness on endothelial expression of vasoactive substances and nitric oxide synthase. Methods Endothelin 1(ET 1),substance P(SP) and nitric oxide synthase expressions of the endothelum in arterioles of hindlimb muscle from control(CON), 4 wk tail suspended(SUS 4),and 1 wk recovered(REC 1) rats were examined by immunohistochemistry technique. Results Our results showed that the total length of arterioles containing nitric oxide synthase(NOS) type Ⅱ and type Ⅲ,ET 1,and SP in gastrocnemius muscle of SUS 4 group were decreased by 11% ( P <0.05),46%,60%,and 27%( P <0.01) respectively,as compared with that of the CON;however,in REC 1,the total length of them were 33%,28%,17% and 77%( P <0.01) longer than those of the CON respectively. In plantaris muscle,the total length of microvessels containing ET 1 and SP were decreased in SUS 4,and were increased in REC 1 as compared with that of the CON. Conclusion The results suggest that the vasoactive substance and NOS expression of endothelium in hindlimb muscles are lowered during simulated weightlessness and heightened during recovery.These changes are suggested to be implicated in one of the important mechanisms accounting for postflight orthostatic intolerance.
