肠道肠球菌AUH-HM195原生质体制备与再生条件初探

摘  要:应用正交试验研究菌龄、酶浓度、酶解时间和渗透压稳定剂种类对肠道肠球菌(Enterococcus hirae)AUH-HM195原生质体制备与再生的影响。结果表明:菌龄与酶浓度的交互作用对原生质体的制备和再生影响最大。最优组合为:菌龄6 h,溶菌酶浓度5 mg/mL,酶解时间1 h,渗透压稳定剂在制备时使用0.7 mol/L KCl,再生时使用17%蔗糖,制备率为93.2%,再生率为14.5%。The effects of bacterial age,enzyme concentration,enzymolysis time and kinds of osmotic pressure stabilizer on protoplast preparation and regeneration of Enterococcus hirae AUH-HM195 were studied by orthogonal test.The results indicated that the interaction between bacterial age and enzyme concentration had obvious impact on protoplast preparation rate and regeneration rate of strain AUH-HM195.The optimal conditions were as follows: bacterial age 6 h,lysozyme concentration 5 mg/mL,enzymolysis time 1 h,osmotic pressure stabilizer 0.7 mol/L KCl for protoplast preparation and 17% sucrose for regeneration.The protoplast preparation rate was 93.2% and regeneration rate was 14.5% under the optimal conditions.
