摘 要:目的比较研究不同类型抗瘤中药制剂对结直肠癌所致NK免疫功能抑制的影响。方法分别制备三氧化二砷(As2O3)、川芎嗪(LHC)、黄芪(AMB)、氧化苦参碱(MOX)、猪苓多糖(PUPS)、蒿甲醚(ART)等6种中药制剂作用后的结直肠癌细胞Colon26再培养上清,以不经中药制剂作用的相应上清为对照;MTT法检测不同中药制剂作用后再培养上清对小鼠脾细胞NK杀伤活性的影响,直接免疫荧光FCM法检测对小鼠脾细胞CD3ε-ζ+表达的影响。结果对NK杀伤抑制的下调,As2O3、PUPS及ART之间未见显著差异(均P>0.05,下调49.65%±8.86%),LHC及AMB之间未见显著差异(P>0.05,下调78.15%±9.81%);AMB的下调作用最强。对CD3ε-ζ+表达抑制的下调,LHC和PUPS的初始下调幅度未见差异(P>0.05,下调88.04%±7.57%),AMB和ART的初始下调幅度未见差异(P>0.05,下调50.00%±8.24%),MOX与其他中药制剂的作用之间均有差异(均P<0.01,完全消除抑制并明显促进表达);LHC的下调作用最强。结论逆转结直肠癌细胞产生的NK免疫功能抑制是中药制剂发挥抗瘤效应的机制之一。临床可考虑联合使用扶正祛邪类及活血化瘀类抗瘤药物组方,以调整患者不利于临床治疗的NK免疫抑制状态。Objective To study the comparative inversion effects of different anti-tumor traditional Chinese medicines on NK immunosuppressions in coloreetal tumor cells. Methods The passage supernatants of Colon 26 ( colorectal tumor cells ) pre-treated were compared with six different traditional Chinese medicines including As2 03 , ligustrazine hydrochloride ( LHC), astragalus mongholicus bge ( AMB), matrine n-oxide ( MOX), polyporus umbellatus polysaechride (PUPS) and artemether (ART) with the corresponding supernatants of Colon 25 which were not treated with any medicines. The effect of passage supernatants pre-treated with different kinds of traditional Chinese medicines on NK killing activity of routine splenocytes was measured by MTT, and CD 3ε^-ζ^+ expression was detected by FCM of direct immunofluorescence . Results There was no difference between the inversion effects of As2O3, PUPS and ART on inhibition of NK killing ( P 〉0.05, down-regulated by 49.65% ± 8.86% ), and between the down-regulating of LHC and AMB( P 〉0.05, down-regulated by 78.15% ±9.81% ) ; AMB had the greatest capability of down-regulation. No difference was found in the initial down-regulation levels of inhibited CD 3ε^-ζ^+ expression between LHC and PUPS ( P 〉 0.05, down-regulated by 88.04% ± 7.57% )and between AMB and ART ( P 〉 0.05, down-regulated by 50.00% ± 8.24% ). But there was difference in the effects between MOX and other traditional Chinese medicines (all P 〈 0.01, inhibition was cleared completely and the expression was significantly elevated); LHC had the greatest capability of down-regulation. Conclusions Reducing NK immunosuppressions produced by colorectal tumor cells should be one of the anti-tumor effects developed by traditional Chinese medicines. Anti-tumor compound such as strengthening healthy and eliminating pathogens and promoting blood circulation can be used clinically to adjust the state of NK immunosuppressions which impede the effect of treatment.
