摘 要:为探讨背根节中神经生长因子(NGF)及trkA蛋白对脊髓可塑性的作用,采用成年大鼠5只,建立单侧备用根模型(即切断一侧L1~4,L6~S1的脊髓节段背根,保留L5为备用根及备用背根节)。术后动物存活5天,灌流固定,取双侧L5背根节(DRG)冰冻切片,用兔抗2.5SNGF及trkA蛋白多克隆抗体分别进行S-P法免疫组化反应,DAB显色。并对免疫反应阳性神经元进行图像分析。其结果手术侧DRG中NGF及trkA蛋白阳性神经元的平均灰度值均分别显著小于非手术侧(P<0.01),表明手术侧阳性神经元中NGF及trkA的含量增加。结果提示。For exploration of the effect of NGF and trkA protein in plasticity of spinal cord, five adult male SD rats were used to establish spared dorsal root preparation(i.e rhizotomy of unilateral L 1~4 , L 5~S 1,hind limb dorsal roots, leaving L 5 as a spared dorsal root). The rats were perfused and fixed with 4% paraformaldehyde after five days. The L 5 dorsal root ganglia were sectioned and 20 um thickness frozen sections were cut. The sections were immunostained with 2.5 s NGF and trkA protein polyclonal antibody respectinely, the S-P kit was used and the sections sere stained with DAB. The average of gray degree of DRG positive neurons was determined with image analysis system. Results: The verage of gray degree in operated side DRGS neurons was significantly lower than that of unoperated side (P<0.01) respectively. It was indicating that the contend of NGF and trkA protein was increased our results suggested that the increase in amount of endogenous NGF and trkA protein expression in spared DRGs neuron might be related with the plasticity after partial rhizothomized dorsal roots.
