摘 要:目的探讨结肠康泰对骨髓间充质干细胞(BMSCs)移植在结肠炎大鼠肠道定植分化的影响。方法雌性SD大鼠以5%2、4、6,三硝基苯磺酸(TNBs)诱导形成结肠炎模型,造模后随机分为模型对照组、BMSCs移植组、柳氮磺胺吡啶(SASP)组、结肠康泰组,第2天将体外分离培养的雄性SD大鼠BMSCs经尾静脉注射到各组雌性大鼠体内。疗程结束后取各组大鼠结肠组织,以Y染色体的性别决定区段sry作为标志,以确定供体BMSCs的植入情况;免疫组化法检测其肠道干细胞标记物Musashi-1的表达。结果经PCR扩增后,凝胶电泳可以检测到结肠组织雄性性别特异性基因区段sry,BMSCs移植组和SASP组阳性率均为20%,结肠康泰组阳性率为30%;免疫组化检测到BMSCs移植组及治疗组大鼠结肠组织Musashi-1均有阳性表达;结肠康泰组与BMSCs移植组比较差异有统计学意义。结论结肠康泰较明显地促进同种异体BMSCs在结肠炎大鼠肠道的定植分化。Objective To explore the effect of jiechang kangtai(JCKT)on the engraftment and differentiation in the intestinal tract of rats with ulcerative colitis(UC)after bone marrow mesenchymal stem cells(BMSCs)transplantation.Methods SD female rats were randomly into model group,BMSCs transplantation group,sulfasalazine salicylazosulfapyridine(SASP)group and JCKT group after the colitis models were induced by trinitrobenzene sulphonic acid(TNBs)at 5% 2,4 and 6.BMSCs separated and cultured from SD male rats in vitro were transplanted into SD female rat models through caudal vein on the next day in each group.At the end of treatment,the colon tissue was collected from rats in each group.The sex-determining gene(sry)located on Y chromosome was taken as the label to confirm the donor's BMSCs transplantation.IHC method was applied to detect the expression of Musashi-1 of its intestinal tract stem cell label.Results After PCR proliferation,the gel electrophoresis was used to detect sry on male gender specific gene segments in colon tissue.The positive rate was 20% in BMSCs transplantation group and SASP group,and was 30% in JCKT group.The positive expression of Musashi-1 in colon tissue of rat was obtained in BMSCs transplantation group and treatment group by IHC detection.There was no significant statistical difference between JCKT group and BMSCs transplantation group in comparison.Conclusion Jiechang kangtai can promote apparently the engraftment and differentiation of BMSCs in the intestinal tract of rats with colitis after allograft of BMSCs.
