摘 要:为了筛选与绵羊羊毛性状相关联的分子标记,探讨Ⅰ型角蛋白中间丝蛋白基因多态位点在不同绵羊品种中的分布规律。采用PCR-RFLP技术对新疆6个绵羊品种Ⅰ型IF基因第1外显子的多态性进行了检测。结果显示,Ⅰ型IF基因第1外显子经MSPI酶切后产生2种等位基因和3种基因型,呈现多态。各绵羊品种之间基因型频率和等位基因频率差异不显著(P>0.05)。和田羊和巴音布鲁克羊群体在该位点上处于Hardy-Weinberg平衡状态(P>0.05)。平均He和PIC分别为0.2374和0.2079。To identify new DNA markers which have significant impact on sheep wool traits and the distribution pattern of polymorphic site of the type Ⅰ IF (intermediate filament wool keratin)gene in different sheep breeds. In this study, the polymorphism of the first exon of Type Ⅰ IF gene was detected by PCR- RFLP method in 6 local sheep breeds in Xinjiang. The results showed that two alleles (M=0. 8609 and N =0. 1391) and three genotypes (MM=0. 7776,MN=0. 1667 and NN=0. 0558) were detected across the 6 sheep breeds. Chi-square analysis indicated that the allele frequency and genotype frequency did not differ among the observed breeds (P〉0.05). This polymorphic locus of Type Ⅰ IF gene was at Hardy-Weinberg equilibrium (P〉0.05) in the Hetian and Bayinbuluke sheep breeds. The average He and PIC were 0. 2374 and 0. 2079, respectively.
