摘 要:目的观察人参皂苷Rg3对大鼠角膜碱烧伤后新生血管形成(CNV)有无抑制作用及其对正常角膜有无毒副作用。方法采用0.5%人参皂苷Rg3滴眼液滴眼21d,通过裂隙灯显微镜、光镜、电镜观察检测人参皂苷Rg3滴眼液的毒副作用。建立角膜碱烧伤后角膜新生血管模型,用0.1%、0.2%、0.5%3种不同浓度人参皂苷Rg3滴眼液及0.1%地塞米松磷酸钠滴眼液4次/d滴眼治疗28d,观察人参皂苷Rg3治疗效果。并用免疫组化方法观察血管内皮细胞生长因子(VEGF)及色素上皮细胞衍生因子(PEDF)表达。结果人参皂苷Rg3滴眼液滴眼21d后无明显不良反应。治疗组与地塞米松组较对照组CNV减少(P<0.01)。治疗组与地塞米松组较对照组VEGF表达降低,PEDF表达增高(P<0.01)。结论人参皂苷Rg3滴眼液局部滴眼无明显毒副作用,且对CNV有显著抑制作用。Objective To investigate the side effects and the effects of the giusenoside Rg3 on corneal neovascularization (CNV) induced by alkali burn in rats. Methods 0.5% ginsenoside Rg3 were used in normal rat cornel for 21 days to observe the side effects with slit lamp , light microscope and electron microscope. Corneal neovascularization was induced in rats by alkali burn which were administed by 0.1%, 0.2%, 0.5% ginsenoside Rg3 and dexamethasone sodium phosphate solution 4times/day for 28days subconjunctivally, corneal angiogenesis was evaluated and observed with slit lamp microscop and electron microscop and VEGF, PEDF level were determined by immunohistochemistry at different interval.Results There were no obviously side effects in rats after the 0.5% ginsenoside Rg3 used for 21days. The area of CNV were decreased than control in ginsenoside Rg3 and dexamethasone group (P 〈0.01 ), VEGF levels were also decreased than control in ginsenoside Rg3 and dexamethasone group( P 〈 0. 01 ) , while PEDF levels were significantly increased than control group. Conclusion Ginsenoside Rg3 could remarkly inhibit CNV in rats and had no side effects.
