摘  要:利用2个株系的4份大豆花叶病毒分离物在低温冰箱(-40℃)、超低温冰箱(-80℃)和液氮(-196℃)3种保存方法下,保存3个月、8个月和15个月,接种感病品种南农1138-2,测定感病后的发病率和病情指数,以寻求适合SMV长期低温保存的方法方差分析结果表明:二指标在保存方法、保存时间、分离物及各种交互作用上均表现显著差异。保存3个月时,3种保存方法均可保持SMV的高发病率和病情指数;保存8个月和15个月时,低温下SMV分离物的发病率和病情指数因来源株系不同而不同,超低温和液氮下的发病率和病情指数均较高,分别可达90.0%和75.0%以上。因此,-80℃超低温和-196℃液氮可以作为SMV较长时期保存的方法,-40℃低温保存通用性差,不适宜长期稳定保存。Plant viruses, such as Soybean Mosaic Virus (SMV) , can live outside the body in room temperature only a very short time, therefore, it is essential to find a way for long term preservation of plant viruses. The present study was to compare different methods of low temperature for long term preservation of SMV. Four isolates of two strains were preserved in low temperature refrigerator ( -40℃ ) , ultra low temperature freezer ( -80 ℃ ) and liquid nitrogen ( -196℃ ) for three, eight and fifteen months. The incidence rate (IR) and disease index (DI) on the susceptible soybean cultivar Nannong 1138-2 inoculated with the isolates were evaluated. The results of ANOVA showed that there were significant differences in IR and DI among preservation methods, preservation periods, isolates and their interactions. SMV isolates preserved in the three methods for three months all kept high IR and DI. The IR and DI of SMV preserved in low temperature refrigerator for eight and fifteen months varied among isolates, but those in ultra low temperature freezer and liquid nitrogen kept more than 90.0% and 75.0% , respectively. Therefore, -80℃ ultra low temperature freezer and -196℃ liquid nitrogen are suitable for preservation of SMV for 15 months, while -40℃ low temperature refrigerator is not a suitable way for long term preservation of SMV due to its fluctuation among isolates. From the high IR and DI values after 15 months preservation under ultra low temperature freezer and liquid nitrogen condition, further longer preservation can be expected, especially for the latter. There-fore, further study is ongoing.
