摘 要:目的研究北五味子Schisandra chinensis(Turcz.)Baill.叶的化学成分及化合物五味子醇甲对脂多糖(LPS)诱导的小胶质细胞激活时所分泌的促炎性因子的影响。方法用色谱法分离并通过波谱分析对化合物的结构进行鉴定;采用LPS诱导的小胶质细胞活化模型,以Griess法检测化合物对LPS激活小胶质细胞的抑制作用;以MTT法检测化合物对小胶质细胞细胞成活率的影响。结果从北五味子叶中分离鉴定了8个化合物:β-谷甾醇(1)、五味子醇乙(2)、五味子乙素(3)、五味子丙素(4)、五味子酯乙(5)、五味子醇甲(6)、(-)-戈米辛M1(7)及(-)-戈米辛L1(8)。五味子醇甲可显著抑制激活的N9小胶质细胞的NO释放。结论首次系统分离并鉴定北五味子叶的化学成分,其中五味子醇甲能明显抑制小胶质细胞的活化。Aim To study the chemical constituent of leaf of Schisandra chinensis(Turcz.) Baill.and the effects of schizandrin on lipopolysaccharide(LPS-induced) proinflammatory cytokine production in activated microglia.Methods Eight known compounds were isolated from the leaf of Schisandra chinensis(Turcz.) Baill.By using LPS-induced microglia activation model,drug screening experiments about schizandrin on anti-microglial cell activity were carried out.By MTT,their effects on the survival rate of microglial cells were detected.Furthermore,inhibition action of the stimulation of LPS on microglial cells was detected by Griess.Results Eight compounds,β-sitosterol(1),gomisin A(2),(±)-γ-schizandrin(3),schisandrin C(4),schisantherin B(5),schizandrin(6),(-)-gomisin M1(7),and(-)-gomisin L1(8),were isolated and identified from the leaf of Schisandra chinensis for the first time.Schizandrin significantly inhibited LPS-induced activation of microglia and production of NO.Conclusion Eight compounds were obtained from leaf of Schisandra chinensis(Turcz.) Bail.for the first time.Schizandrin may be effective on inhibiting microglia activation.
