摘  要:[目的]观察冠通方含药血清对体外培养的人外周血内皮祖细胞(EPCs)数量和功能的影响。[方法]将24只雄性Wistar大鼠随机分为冠通方组(低剂量组、中剂量组、高剂量组)和对照组,制备含药血清和空白血清。采用密度梯度离心法分离获得人外周血单个核细胞,加入各实验血清组培养液培养EPCs,经激光共聚焦显微镜、流式细胞仪鉴定为EPCs,分别在不同作用时间(12h,24h,48h,72h)后,倒置相差显微镜下观察并比较各组EPCs的数量、增殖能力、迁移能力和粘附能力。[结果]与对照组相比,冠通方组可促进外周血EPCs扩增,并显著改善外周血EPCs的粘附、迁移和增殖能力。[结论]冠通方可增加EPC的数量并改善其功能,这可能是冠通方防治冠心病患者经皮冠状动脉腔内成形术(PTCA)加支架植入术后再狭窄的作用机制之一。[Objective] To observe the effect of drug-containing serum in Guantong formula on the number and function of the vitro human peripheral blood endothelial progenitor cells (EPCs). [ Methods ] 24 male Wistar rats were randomly divided into the Guantong formula group (low-dose group, middle-dose group, high-dose group) and the control group, and drug-containing serum and blank serum were prepared. Human peripheral blood mononuclear cells were isolated by density gradient centrifagation, and culture fluid from each experimental serum group was added to cultivate EPCs. The EPCs were identified by laser scanning confocal microscopy and flow cytometry. The number of EPCs, proliferation, migration, and adhesion were observed and compared respectively under the inverted phase contrast microscope in each group at different times (12 h, 24 h, 48 h and 72 h). [Results] Compared with the control group, the Guantong formula group could promote the expansion of peripheral blood EPCs and significantly improve the adhesion, migration and proliferation of EPCs in peripheral blood. [ Conclusion J Guantong formula can increase the number of EPC and improve its function,which may be one of the treatment mechanisms for restenosis prevention in patients with coronary heart disease after percutaneous coronary angioplasty (PTCA) and stent implantation.
