摘  要:目的探讨内皮脂肪酶(EL)在高血压血管重构中的的作用及阿托伐他汀的干预效应。方法用两肾一夹法建立肾性高血压大鼠模型,将21只大鼠随机均分为假手术组(Sham)、肾性高血压组(RH)和阿托伐他汀组(ATV),术后4周末开始用阿托伐他汀30 mg/(kg.d)连续灌胃8周。检测大鼠尾动脉收缩压(SBP),光镜下观察血管结构,并计算胸主动脉中层壁厚与内径之比(MT/LD),免疫组化法检测EL与NF-κB的表达,实时荧光定量RT-PCR检测ELmRNA表达。结果RH组较Sham组SBP、MT/LD、ELmRNA及EL、NF-κB蛋白表达显著升高(P<0.05),而高密度脂蛋白(HDL)、总胆固醇(TC)、低密度脂蛋白(LDL)水平显著降低(P<0.05)。ATV组较RH组MT/LD、ELmRNA及EL、NF-κB蛋白表达显著降低(P<0.05),TC、LDL水平明显下降,HDL水平显著升高(P<0.05)。结论阿托伐他汀可以减轻血管重构,其机制可能与抑制EL表达有关。Objective To investigate the roles of endothelial lipase(EL) in renal hypertensive vascular remodeling and the effects of atorvastatin intervention.Methods Renal hypertension rat model was constructed through two-kidney-one-clip technique.Twenty-one rats were randomly divided equally into sham operation group(sham),renal hypertension group(RH) and atorvastatin-treated group(ATV).Atorvastatin treatment[30 mg/(kg·d)] was started four weeks after model construction and lasted 8 consecutive weeks,given through lavage.All rats were evaluated caudal systolic blood pressure(SBP),microscopic changes in vascular structure and ratio of intima media thickness to lumen diameter(MT/LD) of thoracic aorta.The expressions of EL and nuclear transcription factor-κB(NF-κB) were assessed with immunohistochemical staining and the expression of EL mRNA was tested with real-time quantitative RT-PCR.Results Compared with sham group,RH group showed significant increase in SBP,MT/LD and the expressions of EL,NF-κB protein and EL mRNA,a significant decrease of HDL,TC and LDL level(P0.05).As compared with RH group,MT/LD and the expressions of EL,NF-κB protein and EL mRNA in ATV group were significantly decreased while HDL level significantly increased,TC and LDL levels significantly decreased(P0.05).ConclusionAtorvastatin alleviates the development of vascular remodeling in renal hypertensive rats and the mechanism is potentially associated with decreased expression of EL.
