摘 要:为了确定牛源荚膜血清A型多杀性巴氏杆菌(Pm)的培养特性,本研究采用脑心浸液(BHI)培养基分别于静止、75r/min、250r/min3种培养转速对Pm进行培养,并与加血BHI进行比较,通过绘制生长曲线和小鼠毒力试验对Pm的培养基和培养转速条件进行优化,确定最佳培养条件为,以37℃,0.1%全血脑心浸汤(MB-HI)培养基,75r/min培养16h。将培养的A型Pm灭活后,免疫SPF小鼠,同时设立B型Pm灭活苗免疫组,攻A型Pm后的结果显示,A型灭活菌的保护率为100%,而B型灭活苗免疫组全部死亡。本研究通过对牛源A型Pm培养特性和免疫原性的研究,为新型牛出血性败血症灭活疫苗的研制提供了实验依据。The Pasteurella mutocida capsular serotype A isolated from affected cattle was passaged under optimized cultured conditions of 37 ℃,75 r/min,16 h in MBHI medium and monitored by microscopic examination,multiple PCR amplification,CFU determination and virulence test in mice. No significant changes in bacterial morphology,types-specificity,bacterial density and virulence were observed or detected. The SPF mice were inoculated with inactivated P. mutocida type A,and P. mutocida type B inactivated vaccine was used as a control. The protective rate of the animals by P. mutocida type A was 100%,whereas none of the animals inoculated with P. mutocida type B. were protected. Pathogenicity test showed that P. mutocida type A had no viru-lence to chicken. Therefore the P. mutocida type A could be developed as a potentially novel Haemorrhagic Septicaemia inactivat-ed vaccine Against P. mutocida.
