摘 要:目的:观察CD4+CD25+Foxp3+调节性T细胞、IL-10、TGF-β在哮喘大鼠淋巴液与血液中的水平,以及地塞米松干预的影响,探讨其与哮喘发病的关系。方法:建立大鼠哮喘模型,收集激发后0、24、48 h淋巴液、血液中淋巴细胞,采用流式细胞术(FCM)检测淋巴液及血液中CD4+CD25+Foxp3+T细胞百分率;采用ELISA方法检测血浆及淋巴液中IL-10、TGF-β水平。结果:哮喘组淋巴液和血液的CD4+CD25+Foxp3+T细胞百分率水平、血浆IL-10和TGF-β浓度在各时间点均低于对照组和治疗组(P<0.05);哮喘组淋巴液中CD4+CD25+Foxp3+T细胞百分率水平、IL-10浓度在不同时间点均高于血液水平(P<0.05),而TGF-β浓度则低于血液水平(P<0.05);治疗组淋巴液和血液中CD4+CD25+Foxp3+T细胞百分率水平、血浆IL-10和TGF-β浓度与对照组相比无显著性差异(P>0.05)。结论:哮喘大鼠淋巴液中存在CD4+CD25+Foxp3+调节性T细胞数量以及功能失调,且淋巴液中CD4+CD25+Foxp3+调节性T细胞水平显著高于其血液水平,地塞米松可能通过影响CD4+CD25+Foxp3+调节性T细胞数量以及功能而发挥治疗作用。AIM:To observe the expression of CD4+CD25+Foxp3+Treg,IL-10 and TGF-β in lymph and blood,as well as the effect of dexamethasone on it and investigate the relationship between them.METHODS:Lymph and blood samples of 0 h,24 h,48 h after the last challenge were collected from the rat model of asthma.The percentage of CD4+CD25+Foxp3+Treg were detected by flow cytometer(FCM),while the levels of IL-10 and TGF-β were determined by ELISA.RESULTS:The percentage of CD4+CD25+Foxp3+Treg in the blood and the levels of IL-10 and TGF-β in the plasma in cases of asthma group were significantly lower than that of the control and therapy group at different time points(P0.05);The percentage of CD4+CD25+Foxp3+Treg and the level of IL-10 in lymph were significantly higher than that of blood(P0.05),however,the level of TGF-β in lymph were significantly lower than that of blood(P0.05);There were no significant differences which the percentage of CD4+CD25+Foxp3+Treg in the blood and the levels of IL-10 and TGF-β in the plasma between control group and therapy group(P0.05).CONCLUSION:There is a disbalance both quantity and function of CD4+CD25+Foxp3+ in the lymph of bronchial asthmatic rat,and the percentage of CD4+CD25+Foxp3+Treg in lymph are significantly higher than that of blood,which dexamethasone may effectively increase the percentage of CD4+CD25+Foxp3+Treg in bronchial asthmatic rat.
