摘 要:目的构建骨髓间充质干细胞/骨基质明胶复合体修复大鼠桡骨节段骨缺损,为骨组织工程中制备组织化骨提供理论依据。方法制备成MSCs/BMG复合体和桡骨缺损模型。实验组为A组:MSCs/BMG组;对照组:B组:BMG组;C组:MSCs 1×106/ml组。每组都设置2、4、8、12周四个时间点进行取材。观察术肢X射线放射学、免疫组织化学等检测骨缺损的修复情况。结果X射线放射学评分4、8、12周各组均有明显差异(P<0.01)。Ⅰ型胶原蛋白表达各组随着时间的延长均有明显增强,各组均有明显差异(P<0.01)。结论骨髓间充质干细胞与骨基质明胶复合修复桡骨缺损效果优,可以用于组织化骨。Objective MSCs/BMG composite repair the segmental radial defect in rats with those composites.It may contribut to the development of bone tissue engineering.Methods Prepare MSCs/BMG composite and a bone defect(5 mm in length)was created at the both radial backbone in each rat.The experiment was divided into MSCs/BMG group(A group),BMG group(B group)MSCs group(C group).Morever,each group was subdivided into 4 time points:2,4,8and12weeks with 3 animals at each time point.The activity of operated limb was evaluate by gross observation,radiographic examination and histological analyses were performed to observe the bone defect repair after surgery.Results The X-ray radioactivity grades,4w,8w,and 12w:had the obvious difference with each group(P〈0.01).Ι-collogen protein expression of each group had the obvious enhancement along with the time extension,compared with each other group,the difference was significant(P〈0.01).Conclusion The group MSCs combined with BMG to prepare the tiusse engineering bone is most appropriate for bone tissue engineering.
