摘  要:目的研究久效磷对小鼠的胚胎毒性和致畸性。方法将60只健康SPF级昆明孕鼠随机分为低(0.05mg/kg)、中(0.10mg/kg)、高(0.20mg/kg)剂量久效磷染毒组和对照组(蒸馏水),每组15只。分别于孕期第7~16天(胚胎器官形成期)灌胃染毒,染毒剂量为10ml/kg,每天1次。母鼠于受孕第0、6、12、18、20天称体重。于受孕第20天计数活胎数、死胎数和吸收胎数,并计算活胎率、死胎率和吸收胎率;称量子宫和胎盘重量;观察胎鼠的外观,进行活胎的一般情况检查(体重、身长、尾长)及外观、内脏、骨骼畸形检查。结果在孕期的第0、6天时,各组孕鼠体重间比较,差异均无统计学意义。在孕期的第12、18、20天时,随着久效磷染毒剂量的升高,孕鼠体重呈下降趋势。与对照组相比,高剂量久效磷染毒组活胎数和活胎率均下降,高、中、低剂量久效磷染毒组死胎率和吸收胎率均升高,差异有统计学意义(P<0.05或P<0.01)。随着久效磷染毒剂量的升高,活胎数和活胎率呈下降趋势,死胎率和吸收胎率呈上升趋势。与对照组相比,高剂量久效磷染毒组胎鼠的体重、体长、尾长及胎盘重量均下降,差异有统计学意义(P<0.05)。且随着久效磷染毒剂量的升高,胎鼠的体重、体长、尾长及胎盘重量均呈下降趋势。各组孕鼠子宫重量间比较,差异无统计学意义。各组活胎鼠外观、内脏和骨骼均未发现明显畸形。结论孕期久效磷暴露可致孕鼠吸收胎、死胎发生率升高,久效磷对小鼠具有胚胎毒性。Objective To study the embryotoxicity and teratogenicity of monocrotophos in mice. Methods Sixty pregnant Kunming mice were randomly divided into four groups,15 in each: three groups were exposed to the monocrotophos at the doses of 0.05,0.10 and 0.20 mg/kg, 10 ml/kg through garage, during the period of organ formation (from 7th day to 16th day), once a day and the control group to distilled water. The pregnant mice were weighted in day of 0, 6, 12,18, and 20, and were sacrificed on 20th day of pregnancy. The numbers of living, dead and absorbed fetus were counted, and the uterus and placenta were weighted. The deformity examination was conducted. Results The maternal body weight of exposure groups were lower than those of control group in 12,18 and 20 day of pregnancy. With the increased doses of monocrotophos, the weights of pregnant mice showed a downward trend, Compared with the control group, the rates of dead fetus and absorbed fetus increased significantly, and the rates of living fetus,the fetal body weight and body length,tail length and placental weight decreased significantly (P〈0.05 or P〈0.01) with a dose-dependent manner. Conclusion Monocrotophos exposure during pregnancy may cause an increase of dead fetus and absorbed fetus in mice. Monocrotophos has determinate embryonic toxicity to mice.
