摘 要:目的:观察红体外霉素对体外培养鼻息肉组织中嗜酸性粒细胞趋化因子(eotaxin)的作用。方法:从27例住院病人鼻腔中各取鼻息肉标本一份,将每一份均分4份,分对照组、红霉素组培养1d、3d后。每组各取鼻息肉标本经逆转录聚合酶链式反应(RT-PCR)与免疫组织化学染色检测eotaxin表达。结果:①红霉素组中鼻息肉的eotaxinmRNA光度值较对照组低(p<0.05);②两组鼻息肉中eotaxin蛋白灰度值比较,在培养1d后无明显差异,但培养3d后,红霉素组eotaxin蛋白灰度值较对照组低(p<0.05)。结论:体外红霉素能抑制鼻息肉中eotaxin表达,对鼻息肉的临床治疗有指导作用。Objective: To observe the effect of erythromycin on eotaxin in nasal polyps in vitro. Methods: Each of 27 hospital patients nasal polyp specimens were equally divided into four portions. One group specimens were cultured with erythromycin in RPMI1640, the other group only in RPMI1640 as contral. The specimens in two groups which were used for expriment were taken out at 1 and 3 day after culture. Eotaxin mRNA expression from the two groups above were tested by RT-PCR.Sections from the two groups above were observed under a light microscope by means of immunohistochemical demonstration of eotaxin protein. Results: ①Optical density value of eotaxin mRNA in erythromycin group was lower than contral group(p〈0.05);②There didn't exist difference on gray value of eotaxin protein at the first day afer culture,but gray value of eotaxin protein in erythromycin group was lower than contral group at the third day afer culture (p〈0.05). Conclusion: Erythromycin had inhibition on eotaxin mRNA transcription and protein expression in vitro in nasal polyps,which has directions role in clinic treatment of nasal polyps.
