摘  要:目的研究中药十八反中人参,藜芦合用对CYP1A酶活性的影响,发现基于CYP1A酶活性变化的两者配伍禁忌机制。方法采用高效液相色谱法对人参,藜芦合用对CYP1A酶活性影响进行测定。结果人参与藜芦作用后,大鼠肝微粒体CYP1A的活性呈上升趋势。人参高剂量组、低剂量合用组、藜芦低、高剂量组、高剂量合用组的CYP1A活性上升与空白组比较存在显著性差异(P<0.05),低剂量合用组CYP1A活性发生了极为显著的上升(P<0.01)。结论人参与藜芦配伍前后对CYP1A亚型酶活性调控作用发生明显变化,可能存在基于CYP1A的相反作用,需进一步结合代谢研究加以综合分析,阐明相反作用的机理。Objective To study the mechanism of incompatibility between veratrum and ginsing on the activity of cytochrome P450 1A in the rat liver. Methods Rat liver microsomal cytochrome P450 1A activity was quantitated by high performance liquid chromatography . Results P.gingseng coadministrated with V. nigrum obviously increased the CYP1A activity of rat liver microsomes. Compared with control group, the CYP1A activity was increased in the groups of Veratrum nigrum of lower concentration, Veratrum nigrum of higher concentration and combinative higher concentration(P〈0.05). The group of combinative lower concentration was considerably different from control (P〈0.01),Conclusion The activity of CYP1A was increased in the combinative group. There may exist incompatibility between veratrum and ginseng based on CYP1A. It takes further analysis and research on metabolism to elucidate the drug-drug interaction.
