苦参碱对模型大鼠胃癌前病变的保护作用研究

摘　　要：目的:研究苦参碱对模型大鼠胃癌前病变的保护作用。方法:实验分为正常(自来水)、模型(自来水)、叶酸(40mg·kg-1)、苦参碱(180mg·kg-1)组。灌胃给药,每天1次,持续45d。灌胃结束后分析大鼠体质量变化情况,通过肉眼和显微镜进行病理评分,盲法检测胃组织增殖细胞核抗原(PCNA)、B细胞淋巴瘤/白血病-2(Bcl-2)、干扰素-γ(INF-γ)、白介素-4(IL-4)、血清转化生长因子β(1TGF-β1)的表达水平。结果:苦参碱可以控制萎缩和炎症达到正常(P<0.05),治疗非典型增生(P<0.05),降低PCNA、Bcl-2、INF-γ、TGF-β1表达水平(P<0.05),与叶酸组比较无统计学意义。结论:苦参碱可治疗慢性胃萎缩和通过降低INF-γ的表达来控制胃黏膜炎性细胞浸润;还可通过降低Bcl-2、PCNA、TGF-β1的表达来促进胃癌前病变细胞凋亡,抑制其增殖和胃癌前病变恶化。OBJECTIVE：To study the protection effect of matrine against precancerous lesions of gastric cancer. METHODS：Wistar rats were randomly divided into normal group （water）,model group （water）,folic acid （40 mg·kg-1） group and matrine （180 mg·kg-1） group. Those groups were treated via i.g. gtt once a day for 45 days. The changes of body weight were analyzed and pathological score was carried out by naked eye and microscope. The expression of PCNA,Bcl-2,INF-γ,IL-4,TGF-β1 were detected with blind method. RESULTS：As compared with folic acid group,matrine had sound effect on the control of atrophy and inflammation,atypical hyperplasia treatment,the reduction of expression of PCNA（P〈0.05）. Matrine also can reduced the expression of Bcl-2,INF-γ,TGF-β1（P〈0.05）.There was no statistical significance between folic acid group and matrine group. CONCLUSION：Matrine can be applied to treat chronic gastric atrophy and dintestinal metaplasia,inflammation cell proliferation by reducing the expression of INF-γ. Matrine can also reduce the expression of Bcl-2,PCNA,TGF-β1 to promote precancerous lesions cell apoptosis and inhibit proliferation and deterioration of gastric cancer cell.
