扩展莫尼茨绦虫基因芯片的制作及分析

摘　　要：通过EST计划获得1080个基因片段，制备了扩展莫尼茨绦虫cDNA芯片。该芯片一个域有52行、78列，密度为863点／cm2，其中包含1080个靶基因、9个阳性对照、16个阴性对照、8个空白对照，每个基因做3个重复。用扩展莫尼茨绦虫的成节与头颈节总RNA与芯片进行杂交，获得了成节与头颈节基因表达谱，芯片结果分析差异系数为0．1807，共找到91个在扩展莫尼茨绦虫成节高表达基因，幼节高表达基因81个．其他的基因表达差异不大。通过同源性比对分析，成节高表达基因主要有钙调节蛋白、外致密纤维精子尾、富含半胱氨酸蛋白前体和卵黄蛋白原等；头颈节高表达基因主要有果糖-二磷酸盐醛缩酶、皮层细胞蛋白和胶原等。该芯片可以进一步应用于扩展莫尼茨绦虫基因组等方面的研究，对基因表达差异进行高通量检测。Moniezia expansa cDNA chip was prepared with 1 080 gene fragments from EST program. The chip has a field trip 52 ×78, a density of 863 spots/cm2, which includes 1 080 target genes, 9 positive control, 16 negative control, 8 blank control, three times every genes. Scolex-neck and mature proglottides of M. expansa were selected to conduct two sets of hybridization experiments with the microarray. Gene expression profile was obtained. The dissimilarity coefficient is 0. 1807. High expression genes have 91, has calmodulin, outer dense fiber of sperm tails 1, chorion class high-cysteine protein 12-like precursor, vitellogenin, and so on in M. expansa mature proglottides and the scolex and neck have 81 high expression genes, mainly fructose-bisphosphate aldolase, collagen type XI alpha-2, tegumental protein and collagen triple helix repeat. The newly developed cDNA microarray could be applied in various M. expansa research fields to make the high-throughput detection in the gene expression profiling.
