摘 要:目的在体外研究左氧氟沙星联用利福平对金黄色葡萄球菌防耐药突变浓度(MPC)的影响,为临床合理联用抗菌药物,防止细菌耐药的产生提供理论依据。方法富集浓度为10^10CFU/ml金黄色葡萄球菌ATCC25923菌株,采用琼脂平板二倍稀释法测定左氧氟沙星、利福平单药以及两药以不同浓度联用时对金黄色葡萄球菌ATCC25923的防耐药突变浓度和耐药突变选择窗(MSW)。结果左氧氟沙星、利福平单药对金黄色葡萄球菌ATCC25923的MPC分别为1.0μg/ml和〉480μg/ml,MSW分别为10和〉30000;在联用利福平浓度由0.008~16.38,ug/ml时,左氧氟沙星对ATCC25923的MPC由1.0μg/ml降至0.25μg/ml;在联用左氧氟沙星浓度由0.25~0.80ug/ml时,利福平对ATCC25923的MPC由4.096μg/ml降至0.016μg/ml。结论利福平和左氧氟沙星联合用药可降低各自单药对金黄色葡萄球菌ATCc25923的MPC,缩小MSW。OBJECTIVE To study whether the combination of levofloxacin and rifampicin can influence mutant prevention concentration(MPC) of each single in vitro ,which provide evidence for new strategies for restricting the development of resistance. METHODS We enriehed the cells of 10^10 colony forming units per milliliter Staphylococcus aureus strain ATCC 25923, and used the agar plates dilution method to determine the MPC and mutant selection window MSW of levofloxacin, rifampiein and their combination against the strain ATCC 25923. RESULTS The MPC of levofloxacin or rifampicin alone was 1.0 μg/ml or 〉480μg/ml, and the MSW was 10 and 〉30 000. The MPC of levofloxaein when combined with 0. 008--16. 38 μg/ml rifampiein was decreased from 1.0 μg/ml to 0. 25μg/ml . The MPC of rifampicin when combined with levofloxacin 0. 25 -- 0. 80 μg/ml was decreased from 4. 096 μg/ml to 0. 016μg/ml. CONCLUSIONS The combination of levofloxacin and rifampiein can decrease MPC of S. aureus strain ATCC 25923,and narrow the MSW.
