摘 要:目的:通过对HIF-2a/EPAS1基因多态性与HiHiLo训练前后左心室功能指标进行关联分析,以期从HIF-2a/EPAS1基因多态位点中筛选评价HiHiLo训练中左心室功能效果的分子遗传学标记。方法:对72名健康受试者进行4周的HiHiLo训练,在实验前后测量左心室功能指标;采用PCR-RFLP方法对HIF-2a/EPAS1基因多态位点进行解析;对HIF-2a/EPAS1基因多态性与HiHiLo训练前后左心室功能指标进行关联分析,探寻HiHiLo训练中评价左心室功能效果的分子遗传学标记。结果:150W负荷时,rs13419896EDV变化量存在显著性差异(P<0.05),rs6715787ESV变化量存在显著性差异,EF变化量具有极显著性差异(P<0.01);恢复状态下,rs6715787EF变化量存在极显著性差异。结论:HIF-2a/EP-AS1基因rs13419896多态位点与HiHiLo训练前后150W时EDV相关联,提示可以作为大强度运动时EDV的HiHiLo训练左心室功能效果的分子遗传学标记;HIF-2a/EPAS1基因rs6715787多态位点与HiHiLo训练前后150W时ESV和EF、恢复过程EF相关联,提示可以作为大强度运动时ESV和EF、恢复过程EF的HiHiLo训练左心室功能效果的分子遗传学标记。Purpose:Through making association study between HIF-2a/EPAS1 gene polymorphism and the function indexes of heart which are measured before and after HiHiLo,this paper investigates the molecule genetics markers from the HIF-2a/EPAS1 polymorphic sites.Methods:72 subjects with good health taking part in HiHiLo for 4 weeks,and they are measured cardiac function indexes.HIF-2a/EPAS1 gene polymorphic sites are analyzed by using PCR-RFLP.Association study is made between HIF-2a/EPAS1 gene polymorphism and the function indexes of heart which is measured before and after HiHiLo to find the molecule genetics markers of HiHiLo effect.Result:The rs13419896 polymorphism is associated with the change of EDV at 150W in different genetype.The rs6715787 polymorphism is associated with the change of ESV and EF at 150W in different genetype.The rs13419896 polymorphism is associated with the change of SI at recovery state in different genetype.The rs6715787 polymorphism is associated with the change of EF at recovery state in different genetype.Conclusion:HIF-2a/EPAS1 gene rs13419896 polymorphic site is associated with EDV in 150W,so it may be the molecule genetics markers of EDV.HIF-2a/EPAS1 gene rs6715787 polymorphic site is associated with ESV in 150W and EF in 150W and recovery,and it may be the molecule genetics markers of ESV and EF.
