摘 要:目的探讨改进大鼠Pulsinelli四血管法全脑缺血模型的制作方法。方法健康雄性SD大鼠60只随机分为对照组10只、假手术组10只和模型组40只。模型组大鼠在Pulsinelli四血管闭塞法全脑缺血模型的基础上采用3阶段制作,第1阶段:游离双侧颈总动脉,备线,缝合皮肤;第2阶段:沿第1颈椎内斜向上的神经来寻找翼突孔,用尖端磨得细长的电烙铁分2次灼烧双侧椎动脉;第3阶段:24 h后夹闭颈总动脉10 min。假手术组除不凝闭椎动脉和不夹闭颈总动脉外,其他操作与模型组相同。对照组不做任何处理。结果大鼠造模成功27只,造模成功率为67.5%,模型制作成功的动物存活率为96.3%。苏木素-伊红染色显示对照组大鼠与假手术组大鼠海马CA1区神经元排列整齐,细胞无水肿、变性;模型组大鼠海马CA1区大量神经元变性坏死。结论采用此改进的手术方法可有效提高大鼠Pulsinelli四血管法全脑缺血模型制作成功率和动物存活率。Objective To research the improvement of Pulsinelli four vessels occlusion method in global brain ischemia model of rats. Methods Sixty cases health male SD rats were randomly divided into three groups, the control group (n = 10) ,the sham-operation group( n = 10) and the model group(n = 40). Based on Pulsinelli four vessels occlusion method iu global brain is- chemia model of rats,the making was done by three steps. First,bilateral common cervical artery was dissociated,cervical artery was threaded and the skin was sutured;Seeond,the pterygoid foramen was searched by a nerve coming from atlas,and bilateral vertebral arteries were interdicted with electric iron with a sharp tine;Finally ,'after 24 hours the arteria carotis communis were clamped for 10 minutes. Except the vertebral arteries were not interdicted and the arteria earntis eommunis were not clamped,the operation in the sham-operation group was the same as the mode/ group. Rats in the control group were left intact. Results Twenty-seven rats model wcre successful ,the ratio was 67.5% ,the survival rate of successful rat model was 96.3%. Hematoxylin and eosin coloration showed that hippocampus CAI area neurons of rats were in order,and there were no edema,or denaturalization in the control group and sham-operatinn group;Degeueration and necrosis appeared in the large hippocampus CAI area neurons of rats in model group. Conclusion The modified surgical approach can increase the successful ratio and the survival rates of rat in making the global brain isehemia model with Pulsinelli four vessels occlusion method.
