摘  要:目的探讨钾-氯协同转运蛋白2(potassium-chloride cotransporter-2,KCC2)在耳蜗发育中表达的变化特征及可能的生理学意义。方法取出生后0、3、7、14、21、28、35、60天同窝16只SD大鼠耳蜗组织,每次4只,免疫组化SABC法观察KCC2在不同发育阶段的表达,图像分析系统对螺旋神经节细胞(spiral ganglion neurons,SGNs)阳性表达产物进行差异性灰度分析。结果KCC2在各发育阶段都有表达,以SGNs最明显,毛细胞、支持细胞、外侧壁上也有弱阳性表达。在SGNs的表达结果如下:0、3、7天3个组间无明显差异(P>0.05);14、21、28天3组间表达逐渐下降(P<0.05);28天后各组间无明显差异(P>0.05)。结论①KCC2在耳蜗各发育阶段均有表达;②不同发育阶段,KCC2在SGNs中的表达具有差异,可能对螺旋神经节的正常发育有重要意义;③KCC2在SGNs以外的弱阳性表达,表明其对维持内、外淋巴液中离子平衡具有一定的作用。OBJECTIVE To investigate the expression and the potential physiological significance of potassium-chloride cotransporter-2(KCC2)during the development of rat cochlea.METHODS Cochleae were obtained at the 0、3、7、14、21、28、35、 60th day after birth from the same brood of 16 SD rats.Four cochleae were evaluated at each developmental time point.The exepression of KCC2 were detected by immunohistochemistry and image analysis.RESULTS The expression of KCC2 were detected in all periods after birth,it was located in the cytoplasm and cytomembrane of spiral ganglion neurons(SGNs).There were also weak positive expression in the hair cells、supporting cells and the lateral wall.The quantitative analysis of the SGNs showed that the expression of KCC2 were not significantly different among the groups of 0d、3d、7d(P0.05).The expression were gradually increasing form 14d(P0.05),and reached the topical level at 28d.There were no obvious change after 28d(P0.05).CONCLUSION KCC2 express differently in every stage of developing cochlea and probably play a significant role in the development of the rat cochlea,especially for SGNs.The expression of KCC2 in other positions of cochlea suggest that it may have a certain effect on maintaining the ionic equilibrium of the lymph fluid during the development.
