摘  要:目的评价增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)反义寡核苷酸(antisense oligonucleotide,ASON)和血管内皮生长因子(vascular endothelial growth factor,VEGF)对血管成形术后再狭窄的防治效果。方法28只中国长耳白兔随机分为对照(Ⅰ)组,PCNA ASON(Ⅱ)组,VEGF(Ⅲ)组,PCNA ASON+VEGF(Ⅳ)组,每组7只。分别对血管成形术后再狭窄动物模型进行治疗后,测定各组血管内膜及中膜厚度和面积,对血管狭窄程度进行比较。同时对PCNA蛋白表达进行检测。结果所有实验组均呈不同程度的内膜及血管平滑肌增生,以Ⅰ组最重,Ⅱ、Ⅲ组增生情况较Ⅰ组明显减轻,差异有统计学意义。Ⅳ组与Ⅱ、Ⅲ组间进行秩和检验显示,Ⅳ组增生情况较Ⅱ、Ⅲ组更轻,差异具有统计学意义(P〈0.01),但Ⅱ、Ⅲ组间不具有交互作用,差异无统计学意义(P〉0.05)。Western blotting结果各组间灰度明显不同。结论PCNA ASON与VEGF联合应用显著减轻实验动物动脉成形术后再狭窄,但两者之间无协同作用。Objective To investigate the effect of antisense oligonucleotide (PCNA ASON) and vascular endothelial growth factor (VEGF) gene transduction on restenosis after balloon angiopasty. Methods Chinese rabbits were randomly divided into control group( Ⅰ ), PCNA ASON( Ⅱ ), VEGF only ( Ⅲ ), PCNA ASON + VEGF ( Ⅳ ) groups. Each group included 7 rabbits. Restenosis was evaluated by pathology, immunohistochemistry and Western-blotting for the expression of PCNA, and the depth and area of tunica media and tunica intima were measured. Results All rabbits experienced restenosis on different severities, especially in control group. Lesions were less severe in PCNA ASON and VEGF groups than in controls. The proliferation of smooth muscle and intima significantly ameliorated in PCNA ASON and VEGF combination group than that in PCNA ASON or VEGF groups ( P 〈 0.01 ). But the difference between the PCNA ASON group and VEGF group was not significant (P 〉 0. 7). Conclusion PCNA ASON and VEGF gene transduction are effective in preventing restenosis after balloon angioplasty caused vessel injury in rabbits.
