摘 要:目的分析中国各地HIV-1感染者的vpr基因多态性,比较HIV-1 vpr基因变异的基本特征及其与国外以往研究的异同,为进一步探索HIV-1 vpr的致病机理及可能的基因治疗靶位打下基础。方法提取348例中国各地区HIV-1感染者血浆病毒RNA,运用逆转录聚合酶链式反应(RT—PCR)扩增HIV-1 vpr基因,凝胶电泳鉴定后行基因测序。用Blast、ClustalW、Mega4.0等相关生物分析软件进行vpr基因核苷酸和Vpr氨基酸序列分析,并构建进化树,分析中国HIV.1感染者的vpr基因的多态性、变异特征和亚型的分子流行病学特征。结果按vpr基因分型共发现AB、AE、BC、B、C5种HIV-1亚型,分别占2.60%,55.19%,2.60%,18.18%,21.43%,其中AE亚型比例最大。观察到Vpr氨基酸序列有9个变异率大于20%的位点。结论首次分析中国感染者的HIV-1vpr基因多态性及其基因进化树的特征,发现一些vpr变异率较大的氨基酸位点,这些Vpr氨基酸序列的变异与其致病能力的关系有待进一步研究。Objective To detect and analyze vpr gene diversity of human immunodeficiency virus type 1 ( HIV-1 ) from patients with HIV in China. Different of mutated sites and molecular epidemic characteristics between vpr gene in China and abroad were compared to explore pathogenesis mechanisms of HIV- 1 and possible gene therapy sites in the future. Methods Reverse transcription polymerase chain reaction (.RT-PCR) was used to amplify HIV-1 vpr gene which extracted from plasma of 348 HIV-1 infected individuals. BLAST, Clustal W and Mega biological analysis software were applied to analyze sequences, and identify the characteristics of HIV-1 vpr gene polymorphism and its clinical significance in China. The uprooted tree was calculated according to the neighbor-joining method. Results HIV-1 vpr subtypes in China were CRF01-AE (51.19%), subtypes C (21.43%), subtype B (18. 18%), CRF03-AB (2.60%) and CRF08-BC ( 2. 60% ). Nine sites in vpr deduced amino acid sequences showed more than 20% mutated ratio. Conclusion This paper firstly analyzed HIV-1 vpr gene diversity and the characteristics of vpr gene tree from Chinese patients. Some sites in vpr deduced amino acid sequences showed obvious mutated ratio, which were possibly correlated with clinical manifestation of HIV-1 infected individuals.
