槲皮素对己烯雌酚诱导的仓鼠生精细胞氧化损伤的保护作用

摘　　要：为研究槲皮素(Que)对己烯雌酚(DES)诱导的体外培养仓鼠睾丸生精细胞氧化损伤的保护作用,将生精细胞分别用DES和不同浓度(10、20、30μmol/L)的槲皮素处理,用倒置显微镜观察生精细胞的生长状况,用MTT法测定生精细胞的存活率,用分光光度法检测细胞培养液中超氧化物歧化酶(SOD)、细胞中谷胱甘肽过氧化物酶(GSH-Px)的含量。结果显示,Que显著抑制了DES引起的生精细胞损伤,除低剂量组差异不显著外,中、高剂量组细胞存活率显著上升(P<0.01);Que显著提高了培养液中SOD的活性和细胞中GSH-Px的含量(P<0.01)。说明槲皮素可有效对抗自由基,抑制脂质过氧化,减弱DES导致的生精细胞损伤,提示Que是缓解环境雌激素生殖毒性的有效成分之一。To investigate the protective effects of quercetin on diethylstilbestrol（DES）-induced oxidative damage in cultured hamster spermatogenic cells,the cells were treated with different concentration of DES,and their growth status were observed under the inverted microscope.The viability of the spermatogenic cells was determined by 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyl-tetrazolium bromide（MTT）.The content of superoxide dismutase（SOD） in supernatants and glutathione peroxidase（GSH-Px） in cells was detected with spectrophotography.In result,the quercetin significantly inhibited the DES-induced damage on spermatogenic cells,with the exception of the low-dose group in which no significant difference was observed.The cell survival rate increased significantly in the middle-and high-dose groups.The content of SOD and GSH-Px was significantly increased after medication with the quercetin（P〈0.01）.It was concluded that the quercetin protected the spermatogenic cells against DES-induced oxidative damage through increasing intracellular antioxidants and decreasing lipid peroxidation,indicating that the quercetin played a very important role in ameliorating reproductive toxicity induced by environmental oestrogen.
