摘  要:目的:制备Heymann肾炎精细致病性抗原Gp330并复制人类膜性肾小球肾炎的动物模型。方法:用超速离心和钙沉降方法获取SD大鼠刷状缘微绒毛。用LCA-Sepharose4B亲合层析方法分离纯化Heymann抗原。用多点免疫方法获得抗体并免疫SD大鼠复制Heymann肾炎模型。结果:获取了Heymann肾炎的精细抗原Gp330并成功地用其抗体免疫SD大鼠,使其肾小球基底膜有免疫复合物沉积并产生大量蛋白尿。结论:Gp330是一种复制人类膜性肾小球肾炎模型的新抗原。Purpose To extract nephritogenic Heymann antigen Gp330 and resemble animal model of human membranous nephropathy. Methods By using Ca2+ aggregation of contaminant and differential contrifugation methods, to purfiy the brush border membrance from normal renal proximal tubules of the rats. The nephritogenic Heymann antigen Gp330 was isolated and purified from the extraction by means of LCASepharose 4B affinity chromatography. The rats were immunized with antiGp330 antibody. Results The passive Heymann nephritis which had proteinuria and immune deposits was successfully induced in male SD rats. Conclusions Gp330 is a new antigen which may resemble animal model of human membranous nephropathy.
