摘 要:为了探讨下丘脑多巴胺(DA)能神经元的体外生长发育特性,取新生大鼠下丘脑神经元进行分散培养。用酪氨酸羟化酶(TH)抗血清和免疫细胞化学ABC法显示DA能神经元,并进行图像分析。结果发现,培养1d时即可见TH-免疫反应性(IR)神经元。此后,TH-IR神经元数量增加,迅速生长。至培养7d时,TH-IR神经元的数量、胞体截面积、胞体染色强度和突起长度均达最高峰,且多极神经元所占比例增多,神经元之间建立起多种细胞间接触。培养14d时,TH-IR神经元的胞体大小和突起长度维持在7d时的水平,但数目减少,染色减弱。结果表明,在体外培养条件下,下丘脑DA能神经元的形态和功能在第1周内发育趋于成熟,并可维持到第14d左右。In vitro growth and development of hypothalamic dopaminergic (DA) neurons in the newborn rats was studied by using dispersed neuronal culture, tyrosine hydroxylase (TH) antiserum and immunocytochemical ABC method, and image analysis. TH IR neurons were found as early as in the first day of culture. Thereafter, they were increased in number and rapidly grew. At day 7 in vitro , TH IR neurons reached the highest level of their number, cell body area, staining intensity and process length, respectively. Meanwhile, the percentage of multipolar neurons was increased and intercellular contact established. At day 14, the soma size and process length of TH IR neurons were maintained at the level similar to that at day 7, but their number and staining intensity were decreased. The results indicate that the morphology and function of hypothalamic DA neurons develop to maturation in the first week of culture and maintain the state up to day 14.
