摘 要:目的观察葛根素(puerarin,Pue)对代谢综合征(MetS)-高血压大鼠胰岛素敏感性、血压、肾素-血管紧张素系统以及糖脂代谢的作用。方法:SD大鼠,♀♂各半,除空白对照组食用标准饲料外,其余动物随机喂饲富含高盐(8%)、高脂肪(25%)和高蔗糖(10%)饲料,连续8周;当造模动物出现高血压、糖耐量减退,即建成MetS-高血压病理模型;然后将大鼠随机分为模型对照、模型咪达普利(imidapril,Imid)7mg·kg-1和Pue300,150,75mg·kg-1三个剂量组;分别灌胃给药或蒸馏水,qd×4周后,测定大鼠血压、空腹血糖(FBG)和口服糖耐量试验2小时血糖(OGTT-2hBG)及血浆胰岛素(Fins)、血糖(FPG),并计算胰岛素敏感性(ISI)以及血清甘油三酯(TG)、胆固醇(TC)、肿瘤坏死因子(TNF)-α、内皮素、肾素和血管紧张索含量。结果:与正常对照组比较,MetS大鼠血压升高、FBG和OGTF-2hBG,FPG和Fins含量增高,ISI减弱,血清TGTC,TNF-Ⅱ和肾素、血管紧张素含量增高(P〈0.05或P〈0.01)。经Pue3(Ihng·kg。或Imid处理MetS大鼠后,血压降低(尸〈0.01),FPGFBGOGTr.2hBG降低和IGT改善(P〈0.05或P〈0.01),纠正高胰岛素血症(尸〈O.05或P〈0.01),增强ISI(P〈O.01),降低TG和TC含量(P〈0.05或P〈0.01),下调内皮素、肾素、血管紧张素和TNF—a含量(P〈O05或P〈0.01),上述观察资料,差异均有显著性;而Pue150mg·kg^-1组,对Fins、TNF—α含量和ISI作用,差异则未见显著性(P〉0.05);Pue75mg·kg^-1组对上述各指标,差异均未见显著性(P〉0.05)。结论:Pue或Imid对MetS大鼠,能对抗胰岛素抵抗、增强胰岛素敏感性、纠正高胰岛素血症,降低血糖和血脂,拮抗内皮素、肾素.血管紧张素系统,降低血压。葛根素以上效应呈量效关系。Objective To investigate the effects and effective mechanisms of puerarin(Pue)on insulin resistance, hypertension, rennin-angiotensin system, vasoactive substance and blood glucose and lipid in metabolic syndromic(MetS)-hypertensive(Ht) rat fed high salt-fat-sucrose-diet. Methods Male and female for half and half Sprague-Dawley(SD)rats were randomized to receive feeding a high salt (8%)-fat(25%)-sucrose (10%)-enriched diet qd for 8 weeks, MetS-Ht rat model that the rats were exposed hypertension and impaired glucose tolerance (IGT) essentially established; and then the rats were randomly divided into model control group, imidapril (Imid) group 20mg·kg-1, and Pue 300mg·kg-1, 150mg·kg-1, 75mg·kg-1, except normal group fed standard rat chow; solvent or the drugs were adiministered by intragastric administration (ig) qd for 4 weeks. The blood pressure(BP), fasting plasma glucose(FPG) and fasting insulin(Fins)as well as insulin sensitivity index(ISI)were be measured; fasting blood glucose (FBG), 2-hours blood glucose (2hBG) after oral glucose tolerance test(OGTT), the contents of triglyceride (TG), total cholesterol (TC), rennin, angiotensin, endothelin(ET), tumor necrosis factor (TNF)-α were determined after treatment. Results In MetS-Ht rats, the BP were raised, FBG and OGTT-2hBG, FPG and Fins were increased, ISI weakened, the concentrations of TG, TC, rennin, angiotensin, TNF-α were increased compared with the normal control rat(sP0.05 or P0.01). BP were decrease (P0.01), FPG, FBG, OGTT-2hBG lowered and IGT improved (P0.05 or P0.01), hyperinsulinemia moderated (P0.05 or P0.01), ISI strengthened(P0.01), contents of TG and TC reduced (P0.05 or P0.01), concentrations of rennin, angiotensin, ET and TNF-α downregulated (P0.05 or P0.01), all above data in the MetS-Ht rats treated by Imid or Pue 300mg·kg-1 were significant difference. And there had no significant difference in concentrations of Fins and
