摘 要:目的探讨淋巴细胞趋化因子(lymphotactin,Ltn)与大鼠白色念珠菌肺炎发生发展过程的相关性。方法选用清洁级健康雄性SD大鼠(150~180g,鼠龄30~40d)80只,随机分为模型组与对照组,每组40只。实验组从气管注入白色念珠菌菌液制备白色念珠菌肺炎模型,分别在接种完成后1d、3d、7d、11d处死大鼠(各组10只),取心室血,采用双抗体夹心ABC-酶联免疫吸附试验法测定血清Ltn、γ干扰素(IFN-7)、白介素2(Ib2)水平,取肺组织行组织病理检查及病原体培养计数,应用SPSS13.0统计软件对数据结果进行处理。结果模型1d、3d、7d、11d组均见明显炎症反应,Szapiel评分分别为(1.70±0.48)分、(3.60±0.70)分、(2.90±0.57)分、(1.30±0.52)分,菌体负荷数分别为(3.50±1.07)×10^6cfu/ml、(4.60±1.08)×10^5cfu/ml、(1.30±0.48)×10^5cfu/ml、(0.10±0.32)×10^6du/ml。模型1d、3d、7d组Ltn、IFN-γ、IL-2水平均高于对照组(P〈0.05),模型组Ltn的消长与炎性程度、菌落数、IFN-γ、IL-2水平呈正相关(7=0.816、0.647、0.846、0.823,P值均〈0.05)。结论Ltn参与了真菌感染过程,与IFN-γ、IL-2在消除病原体方面起着相互协同的积极作用。Objective To investigate the correlation between lymphotactin (Ltn) and pulmonary candidiasis albicans in rats. Methods Eighty male SD rats (150-180 g,30-40 days of age) were randomly divided into control group and model group ( n = 40). In model group, pulmonary candidiasis albieans model was prepared by the injection of Candida albicans from the trachea. Experimental animals were sacrificed at 1 d, 3 d, 7 d, 11 d after vaccination, and ventricle blood was taken. The levels of Ltn, interferon-γ (IFN-γ),interleukin-2 (IL 2) in serum were measured by double-antibody sandwich ABC- enzyme linked immunosorbent assay. Lung tissues were taken for tissue pathology examination and pathogen count training. The data were analyzed by SPSS 13.0. Results Inflammatory reaction was observed in model 1 d,3 d,7 d, 11 d groups. Szapiel scores were 1.70±0.48,3.60± 0.70,2.90±0.57, 1.304±0.52, respectively. The numbers of bacterial load were (3.50±1.07)×10^6 cfu/ml, (4. 60±1. 08)×10^6 cfu/ml,(1. 30±0.48)×10^6 efu/ml, (0.10± 0.32)×10^6 cfu/ml, respectively. Compared with control group, the levels of Ltn, IFN-γ and IL-2 increased in model 1 d, 3 d, 7 d, 11 d groups ( P 〈 0.05). There was positive correlation between Ltn level and the degree of inflammation, the number of coenobium,the levels of IFN-γ and IL-2 in model group ( r =0. 816,0. 647,0. 846,0. 823, all P 〈0.05). Conclusions Ltn might participate in the course of fungal infection, and play a positively synergistieal role with IFN-γ, IL-2 on the elimination of pathogens.
