摘 要:为探讨一氧化氮(NO)在听源性惊厥点燃中的可能作用,用NADPH-黄递酶组织化学方法和Fos免疫细胞化学方法,研究了听源性惊厥易感大鼠惊厥和点燃后听觉核团和前脑结构内NOS-Fos双重阳性神经元的分布及差异。结果显示:一次惊厥后,下丘内双重阳性神经元较多,近70%~80%的NOS阳性神经元呈Fos阳性,其他听觉核团和前脑结构内仅见少量NOS-Fos双重阳性神经元。点燃后,听觉核团和前脑结构内NOS-Fos双重阳性神经元明显增加,而且嗅周皮质第Ⅱ层浅部多数NOS弱阳性神经元呈Fos阳性,第Ⅲ层出现大量NOS-Fos双重阳性神经元。上述结果表明,听源性惊厥点燃诱导NOS表达增加。In order to explore the role of nitric oxide (NO) in audiogenic kindling,the present study investigated the distribution and difference of NOS Fos double labelled neurons in the auditory nuclei and forebrain structures after single (AS) and kindled (KAS) audiogenic seizures in P77PMC rats,a strain of Wistar audiogenic seizures prone rats.Using histochemical and immunocytochemical methods,we obtained the following results:(1) in AS rats,a few of NOS Fos double labelled neurons distributed in the auditory nuclei and forebrain structures.However,there were a sum of double labelled neurons in the inferior colliculus,and approximately 70%~80% NOS positive neurons were also Fos positive.(2)in KAS rats,NOS Fos double labelled neurons significantly increased in all of the auditory nuclei and forebrain structures( P <0 01),expecially in the perirhinal cortex,a lot of double labelled neurons appeared in the deep layer of the perirhinal cortex,and almost all of NOS positive neurons were also Fos positive.These results indicated that audiogenic kindling could increase NOS expression,and the increased expression of NOS might play a role in sustaining the increased neuronal susceptivity.
