摘  要:为了探讨提高小鼠卵细胞质内单精子注入受精率的方法,选取鼠龄12~14周的健康昆明白小鼠作为精子和卵子的供体,采用ICSI技术,以受精后二细胞卵裂的形成率为指标,了解不同采卵时间、不同微注射针参数及不同培养液对细胞质内单精子注入的影响。结果表明,hCG注射后18~19h采卵,用针尖内径为4~5μm、斜面角度为35~40度的微注射针进行ICSI操作,受精后卵子置CZB中培养可获得较多的2细胞胚,卵裂率明显高于其余各组(28.10%),有高度显著性差异(P<0.01)。结果提示:恰当的采卵时间。To study methods raising average percentage of fertilization by intracytoplasmic sperm injection (ICSI) in muose eggs. Methods: Kunming white mice (12 14 weeks of age) were use as donors of sperms or eggs. The presentation of 2 cell blastomere was considered a success of fertilization. The influence of different parameter of micropipette,phases of collecting eggs and media on ICSI was investigated. Results: The results showed that the average percentage of 2 cell stage was clearly higher (28 10%) than that of other groups while using micropipette tip with internal diameter 4 5 μm and angle of incline plane 35 40, collecting eggs on 18 19 h after injecting human chorionic gonadotrophin、 application of Chatot Ziomek Bavister medium. Conclusion: Results suggest that proper parameter of micropipette,phases of collecting eggs and selected medium were closely related to percentage of fertilization during ICSI course.
