摘 要:【目的】建立新疆紫草组培快繁技术体系。【方法】以新疆紫草无菌苗的茎尖为外植体,筛选适合茎尖萌发、增殖、生根的激素组合。【结果】茎尖最佳萌发培养基为MS+TDZ 1.0 mg/L,其萌发率为100%。芽增殖培养基为MS+TDZ 1.0 mg/L,芽增殖倍数为13.50倍,继代壮苗培养基为MS+NAA 0.3 mg/L+6-BA 0.5mg/L,生根培养基为MS+NAA 0.1 mg/L,生根率为84%。降低6-BA浓度到0.05 mg/L可大幅降低玻璃化发生率。经过此培养基继代后,再在原生根配方中生根,生根率提高到95.32%。【结论】茎尖是新疆紫草快繁的最佳外植体。[Objective]The aim of the study was to establish rapid propagation system of Arnebia euchroma (Royle.) Johnst. [ Method ] The proper combination of growth regulator was screened by using the stem - tip of Arnebia euchroma (Royle) as a explant, proliferate and root - age. [ Result ] The results showed that the optimal germination medium was MS + TDZ 1.0 mg/L, whose germination rate was 100% and the bud multiplication medium was MS + TDZ 1.0 mg/L with 13.50 folds and successive healthy seedling medium was MS + NAA 0.3 mg/L + 6 - BA 0.5 mg/L, the best rooting medium was MS + NAA 0.1 mg/L, the rate of rooting was 84 %. Where the consentration of 6 - BA decreased by 0.05, the rate of vitrification could be largely decreased and when cultured in this medium, the rooting rate could increased to 95.32 %. [ Conclusion ] Stem tip is the best explant of Arnebia euchroma (Royle.) Johnst rapid propagation .
