摘 要:通过研究1型糖尿病(T1DM)大鼠肾皮质和正常大鼠肾皮质蛋白质表达的差异,进一步探讨T1DM引起肾脏病变的可能机制。实验采用链脲佐菌素(STZ)诱发SD大鼠产生高血糖,建立T1DM模型,应用双向凝胶电泳技术分离大鼠肾皮质蛋白。电泳图谱结果:正常组分辨出蛋白斑点468±34个;DM组分辨出蛋白斑点423±23个,其中差异显著的蛋白斑点43个,与正常组比较,DM组有15个蛋白斑点表达上调,24个表达下调,2个失表达,2个蛋白斑点只在DM组表达。本实验证明了T1DM大鼠肾皮质蛋白质表达发生了显著变化,差异蛋白质点的进一步鉴别研究将有助于探讨T1DM肾病的发病机制。To explore the possible mechanism of nephropathy of type 1 diabetes mellitus (T1DM), the protein difference in renal cortex between diabetic rats and normal rats was examined. T1DM model was successfully obtained by streptozocin (STZ) injection to established high level of blood glucose in SD rats. Then the proteins of renal cortex were separated by two-dimensional gel eleetrophoresis (2- DE). The results showed that about 468±34 and 423± 23 protein spots were detected in the maps of control and DM groups respectively. There are 43 protein spots providing significant differential expression, of which 2 protein spots are only expressed in control rats, and 2 protein spots in DM rats, 15 up-regulated spots in diabetic rats expression, and 24 down-regulated in diabetic rats tissue. It is clearly seen that a significant difference was observed between diabetic rat renal cortex and control rat in protein expression, which contribute to further research of the pathogenesis of type 1 diabetic nephropathy.
