摘 要:为了研究高效液相色谱法分析测定金铁锁多糖中主要所含单糖的组成及其比例,将多糖样品加硫酸水解,1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化后用HPLC-DAD进行分析测定,并将单糖对照品同法衍生化后测定得到各单糖衍生化物的标准曲线,对样品水解液中主要所含单糖进行测定分析。结果显示14.85 mg金铁锁多糖中含有半乳糖(5.02 mg)、葡萄糖(2.51 mg)、木糖(1.20 mg)、鼠李糖(0.31 mg),其摩尔比为15∶7∶4∶1。实验表明该分析测定方法灵敏、准确、可行。To analyze the components of monosaccharides in Psammosilene tunicoides Polysaccharide by HPLC- DAD, the sample was first hydrolyzed with sulfuric acid, and then neutralized with barium carbonate to obtain a mixed monosaccharide solution. The obtained monosaccharides and the reference substances were derivatized with PMP(1 - phenyl- 3 - methyt7 5 - pyrazolone) under alkaline condition and the redundant PMP was removed with chloroform. The analysis was .carried out by HPLC-DAD. The flow phase were ammonium acetate (A) and acetonitrile(B), with B kept 16% from 0 to 13 minutes, 16% changed to 21% from 13 to 14 minutes and kept 21% from 14 to 30 minutes. The results showed that 14. 85 mg Psamrnosilene tunicoides Polysaccharide contains 5.02 mg galactose, 2. 51 rng glucose, 1.20 rng xylose and 0. 31 mg rhamnose. The molar ratio is 15 : 7 : 4 : 1. It is simple, sensitive and accurate to analyze and to determine the contents of monosaccharides in polysaccharide after derivatization with PMP by HPLC-DAD.
