摘  要:平48(以下简称P48)是通过转育方法将性连锁平衡致死系的群体性比控制基因转移到白云品系培育而成的。研究以P48与白云(BY)这对近等位基因系为分析材料,利用AFLP筛选并分析差显标记,经PAGE电泳、片段回收、克隆、测序及与家蚕(P50)基因组数据库序列比对发现家蚕性连锁平衡致死系Z染色体特异性连锁标记C094,该标记与P50的对应序列间存在明显差异。进一步设计差异引物对P48与P50及其杂交F1代进行PCR验证,确定C094标记与家蚕性连锁平衡致死系P48致死基因l1连锁,这为进一步鉴定及克隆致死相关基因奠定了基础。Silkworm Ping 48 strain ( P48 in short) was obtained by transforming of group sex ratio-controlled letal genes into normal strain Baiyun, which was a near isogenic line of P48 strain. P48 and Baiyun strains were analyzed by amplification fragment length polymorphism (AFLP) in present paper for screening the Z chromosome markers in silkworm sex-linked balanced lethal strain. C094, one Z linked marker, was found by DNA retrieving, cloning, sequencing, and blasting silkworm (P50) whole genome database on NCBI. PCR primers were designed to perform the difference identification among P48, PSO and their F1 hybridization. The results showed that C094 marker was genetically linked with l1 lethal gene in silkworm balanced sex-linked lethal strain. This benefits the characterizing and cloning of lethal related genes in silkworm sex-linked balanced lethal strain.
