摘  要:首次采用特殊方法从感染兔红细胞上分离、纯化两种血吸虫组分抗原SjR47和SjR12,实验结果证明,前者属卵源性抗原,后者为虫源性抗原.两次实验结果均表明,两种组分抗原能诱导出明显的抗病免疫力,实验1获得54.95%-72.36%的肝内减卵率,实验2获得52.57%-57.27%的肝内减卵率和50.73%-74.70%的成熟卵减少率,并均可使肝内肉芽肿病变明显减小,尤以SjR47效果显著.两种抗原诱导的抗肝内肉芽肿保护性免疫调节机制的研究结果表明,两者均能诱导小鼠产生高水平的IgG,测定IgG及其亚类动态显示,在感染后0-12wk时,免疫小鼠血清IgG和IgG_1水平均显著高于佐剂和感染对照组者(P<0.01),特别是能反映保护性免疫力指标的IgG_1升高非常明显:SjR47免疫鼠的脾T淋巴细胞亚群表型动态研究显示,SjR47免疫鼠的CD4~+和CD8~+T细胞在感染4wk时均升高,随后下降.CD4~+/CD8~+比值也随时间延长而下降,呈现免疫下调,其变化趋势同肉芽肿病变减小相一致;SjR47免疫鼠的细胞因子水平动态观察结果表明,SjR47免疫鼠后可使TNF-α在小鼠体内的活性水平较平显著上升(成虫期前),减弱了TNF-α对虫卵肉芽肿形成的介导作用以及对雌虫排卵的促进作用,导致肝内肉芽肿病变减小和虫卵减少。本研究结果表明。SjR47 and SjR12 fractional antigens on erythrocytes of rabbits infected with Schistosomajaponicum were successfully isolated and purified by a special method. Charaterization study by IFA indicated that these antigens shared egg and worm origin repectively. Further study demonstrated that the effects of anti-pathology immunity could be induced by the two antigens in mice. Results of 2 immunization tests showed that the reduction rates of total eggs in liver of immunized mice were 54. 95%-72. 36% in experiment 1, and the reduction rates oftotal eggs and mature eggs were 52. 57%-57. 25% and 50. 73%-74. 70% respectively in experiment 2. The egg granulomatous formations in immunized mice were all diminished in twoexperiments. We further studied the mechanism of protective immunoregulation of egg gramulomatous formation in mice immunized with the two antigens. It was demonstrated thatthe specific and subclass antibody levels of sera from immunized mice were significantly higher than those of the infection controls, particanly the IgGl, in 0-12wk after infection;Thelevels of splenic CD4^+ and CD8^+ T cells in immunized mice were high in the early stage(4wk) post infection and then decreased gradually, the ratio of CD4^+/CD8^+ T cells was alsodecreased gradually. The results indicated that the cellular immune response may be inhibited in immunized mice F Serum TNF-α levels of immunized mice were-found significantly higher than those of infection controls in the early stage (before worm maturing) post infection,suggesting that this cytokine may mediate egg granuloma formation and induce a reducedegg-laying capacity of the parasite. These results indicated that the protective immunity ofanti-pathology in mice immunized with the two antigens appeared predominantly in earlystage after infection and closely related to cellular (cytokine) as well as humoral immunity.
