丹参酮ⅡA对马兜铃酸肾病肾间质微血管病变的作用

摘  要:目的研究丹参酮ⅡA(TSN)对马兜铃酸肾病(AAN)肾间质微血管(PTCs)病变的作用。方法将30只雌性Wister大鼠分为3组:模型组和治疗组各10只,灌胃关木通水煎剂,对照组10只,灌胃饮用水。2h后,治疗组给予丹参酮ⅡA腹腔注射,对照组和模型组注射生理盐水。实验第12周处死全部大鼠,留取血、尿和肾组织标本,分别作生化、病理、免疫组化、免疫印迹、逆转录聚合酶链反应等检查。结果与模型组相比,治疗组大鼠肾组织PTCs密度增加,低氧诱导因子-1α(HIF-1α)表达减少,肾功能损害减轻,尿蛋白减少(P均<0.01)。结论TSN能改善肾脏PTCs病变,保护肾功能,延缓AAN进展。Objective To investigate effect of tanshinone ⅡA(TSN) on impairment of peritubular capillaries(PTCs) in rats of aristolochic acid nephropathy(AAN).Methods Thirty rats were divided into 3 groups:control,model and treatment groups.The rats in model and treatment groups were infused with caulis aristolochiae manshuriensis decoction.2 h later,the rats in treatment group was injected with tanshinone ⅡA.Serum creatinie(Scr),blood urea nitrogen(BUN) and urine protein were detected at the end of study.HE and Masson stainings were used to observe renal pathology.Immunohistochemistry were used to detect expression of CD34.Expression of hypoxia-inducible factor-1α(HIF-1α) were determined by western blot and RT-PCR.Results Compared with model group,density of PTCs increased more,but expression of HIF-1α,urinary protein,Scr and BUN were significantly decreased in treatment group(all of P values were less than 0.01).Conclusion TSN can improve PTCs injury,protect renal function and slow progression of AAN.
