摘 要:目的:探讨低氧、运动对骨骼肌蛋白质合成的作用及其机理。方法:雄性SD大鼠随机分为对照组、低氧组、运动组、低氧运动组,每组各20只。实验第1、28 d分别取材,放免法测试骨骼肌睾酮含量,荧光定量PCR测试ARmRNA表达,western blot检测AR蛋白表达,EMSA测试AR与DNA结合能力,BCA法测试骨骼肌总蛋白质含量。结果:1)28 d运动组比对照骨骼肌睾酮含量下降,ARmRNA和AR蛋白表达升高,AR与DNA结合能力明显升高,有非常显著性差异,骨骼肌总蛋白质含量升高,有显著性差异。2)28 d低氧组比对照组骨骼肌睾酮含量下降,有显著性差异,ARmRNA升高,AR蛋白表达降低,AR活性升高,骨骼肌蛋白质含量升高。3)28 d低氧运动组比运动组骨骼肌睾酮含量明显降低,ARmRNA、AR蛋白表达、AR活性和骨骼肌总蛋白含量下降,均有显著性差异。结论:1)运动后进行低氧暴露比单纯运动可能通过睾酮-AR含量-AR活性各水平抑制蛋白质合成。2)单纯低氧暴露、运动或运动后进行低氧暴露通过睾酮水平调节AR,最终影响骨骼肌蛋白含量。Purpose To study the effect and mechanism of hypoxia and exercise on muscle protein synthesis under simulated "Living high-training low(Hi-Lo)"condition.Methods Male Sprague-Dawley rats were randomly divided into four groups as follow :Control group,Hypoxic group,Exercise group,Hypoxic exercise group with 20 rats in each group.In the 1st、28thday of test,the samples were collected.The concentration of muscle testosterone were tested by radioimmunoassay,AR gene and protein expression were detected by real-time fluorescent quantitative PCR and western blot analysis respectively,AR and DNA binding ability was measured with EMSA,muscle protein content was assessed with BCA.Results 1) After 28 days,compared with control group,the exercise group's muscle testosterone content decreased,ARmRNA and AR protein expression increased,AR and DNA binding ability increased significantly,muscle total protein content enhanced significantly.2) After 28 days,compared with control group,the hypoxic group's muscle testosterone content respectively decreased significantly,ARmRNA improved,AR protein expression decreased,AR activity increased much,muscle total protein content enhanced.3) After 28 days,compared with exercise group,Hypoxic exercise group muscle testosterone content decreased,ARmRNA decreased,both AR protein expression and AR activity decreased significantly,muscle total protein content decreased significantly.Conclusions.1) Hypoxia after exercise inhibited muscle protein synthesis much via each leval of testosterone、AR content、AR activity.2) Hypoxic exposureexercise、Hypoxia after exercise regulated AR by changing muscle testosterone leval,and infulenced muscle protein content finally.
