人参三七组方对人脐静脉内皮细胞血管内皮生长因子分泌及其受体2表达的影响

摘　　要：目的:探讨益气活血中药人参三七组方对人脐静脉内皮细胞(human umbilical vein endothelial cell,HUVEC)血管内皮生长因子(vascular endothelial growth factor,VEGF)及其受体2(vascular endothelialgrowth factor receptor-2,VEGFR-2)表达的影响。方法:体外培养HUVEC,分为高剂量人参三七组方(0.4 mg/mL)组、中剂量人参三七组方(0.2 mg/mL)组、低剂量人参三七组方(0.1 mg/mL)组,另设碱性成纤维细胞生长因子(bovine basic fibroblast growth factor,bFGF,320 U/mL)阳性对照组和只加培养液的空白对照组。酶联免疫吸附测定法检测细胞上清液中的VEGF含量,免疫细胞化学法检测VEGFR-2阳性细胞数及其光密度值,蛋白印迹法检测VEGFR-2蛋白的表达。结果:与空白对照组比较,高剂量人参三七组方组和bFGF组细胞上清液中VEGF含量增加(P<0.05),VEGFR-2阳性细胞数及光密度值增加,细胞中VEGFR-2蛋白表达增强。结论:促进HUVEC分泌VEGF和表达VEGFR-2可能是人参三七组方促血管生成的基础。Objective：To investigate the effects of Radix Ginseng and Radix Notoginseng formula on secretion of vascular endothelial growth factor（VEGF） and expression of vascular endothelial growth factor receptor-2 （VEGFR-2） in human umbilical vein endothelial cells（HUVECs） in vitro. Methods：HUVECs were cultured in vitro.Bovine basic fibroblast growth factor（bFGF） at concentration of 320 U/mL and Radix Ginseng and Radix Notoginseng formula at concentrations of 0.1,0.2 and 0.4 mg/mL were used to culture with HUVECs.And HUVECs in blank control group were cultured with culture solution only.After 24-hour culture,the content of VEGF in supernatant was detected by enzyme-linked immunosorbent assay and the expression of VEGFR-2 was detected by immunocytochemical staining and Western-blotting. Results：Radix Ginseng and Radix Notoginseng formula at 0.4 mg/mL,the same as bFGF,increased VEGF content in the HUVEC supernatant and the number of VEGFR-2-positive HUVECs.Expression of VEGFR-2 protein in high-dose Radix Ginseng and Radix Notoginseng formula group was up-regulated as compared with the blank control group. Conclusion：Radix Ginseng and Radix Notoginseng formula can promote HUVEC proliferation and secretion of VEGF,as well as the expression of VEGFR-2 protein,which may be one of the mechanisms of Radix Ginseng and Radix Notoginseng formula in promoting angiogenesis.
