大鼠静脉注射乳化异氟烷的急性毒性

摘　　要：目的研究大鼠尾静脉注射乳化异氟烷的急性毒性并确定其半数致死剂量（ID50）。方法清洁级SD大鼠60只，按雌雄分层随机分为6组，溶媒对照组（C组）和不同剂量乳化异氟烷组（I1-5组），每组10只，雌雄各半。I1-5组分别单次尾静脉注射乳化异氟烷225．6、192．0、163．2、139．2、118．8mg／kg，以心电图成一直线判断死亡，采用Bliss法计算乳化异氟烷的LD50及其95％可信区间。C组单次尾静脉注射等容积30％脂肪乳。注射乳化异氟烷后观察2周。结果大鼠单次尾静脉注射乳化异氟烷后出现翻正反射消失、呼吸抑制、心律失常或死亡；乳化异氟烷的LD50为170．76mg／kg，95％可信区间为153．00～190．56mg／kg。结论大鼠单次尾静脉注射乳化异氟烷的LD50为170．76mg／kg，急性毒性主要表现为呼吸循环抑制。Objective To investigate the acute toxicity of intravenous isoflurane and determine the median lethal dose （ LD50 ） in rats. Methods Sixty SD rats were randomly divided into control group （ n = 10 ） and different doses of isoflurane group （I1-5 group, n = 10, 5 male, 5 female）. The I1-5 group received a bolus of intravenous isoflurane 225.6, 192.0, 163.2, 139.2 and 118.8 mg/kg injected via tail vein over 10 seconds respectively and LD50 was calculated by Bliss. In control group 30% lipid emulsion was administered instead of isoflurane. The survived rats were observed for two weeks. Results The animals exhibited loss of righting reflex, cardio-respiratory depression or death. The LD50 of intravenous isoflurane was 170.76 mg/kg and 95 % confidence interval was 153.00-190.56 mg/kg. Conclusion The LD50 of intravenous isoflurane was 170.76 mg/kg, and the acute toxicity of intravenous isoflurane is cardio-respiratory depression.
