摘　　要：老舍有着怎样的文化形象和价值意义？过去的文学史一般多从＂市民＂、＂京味＂和＂幽默＂等加以概括,忽略了他的精神世界和文化价值的独特性。又时常理所当然地从鲁迅所开启的现代启蒙主义传统来审视他的文学创作,使他陷入＂被别人表述＂的阐释困境。论文提出应该回到老舍那里去,寻找老舍文学的独特性,认为老舍是现代中国最伟大的伦理型作家,具有丰厚而深沉的伦理情怀;伦理诉求是老舍文学创作别具一格的地方;社会正义与个人德性是他的小说最为集中表达的伦理观念。Basing on isolation and identification of subgroup J avian leukosis virus,a DNA fragment of 921 bp encoding gp85 of subgroup J avian leukosis virus was amplified by PCR.The gene was then cloned into pET28a expression plasmid.After sequencing and alignment with the standard strain,the recombined plasmid was transformed into E.coli BL21（DE3） and induced by IPTG.SDS-PAGE analysis showed that the fusion protein of gp85 was efficiently expressed and the protein size was about 40 KD.The result from Western blot showed that the fusion protein of gp85 could react specifically with the ALV-J positive serum.The purified recombinant protein of gp85 was used as coating antigen for the development of an ELISA assay to detect specific antibodies against subgroup J avian leukosis virus.The experimental conditions of ELISA were explored and determined.Clinical samples were detected by the established ELISA.The result showed that the recombinant protein gp85 could be used as antigens in an ELISA method to detect specific antibodies against subgroup J avian leukosis virus.
