CXCR4在神经母细胞瘤细胞SK-N-SH上的表达及其对化疗敏感性研究

摘　　要：目的:探讨CXCR4在神经母细胞瘤细胞SK-N-SH表面的表达及其对化疗药物环磷酰胺(CTX)、吡柔比星(THP)的敏感性。方法:体外培养SK-N-SH神经母细胞瘤细胞,流式细胞仪测定CXCR4在其表面的表达。通过CH50试验明确化疗药物CTX、THP对SK-N-SH细胞的效应剂量。最后通过Real-time PCR及流式细胞术在基因及蛋白水平检测化疗药物对SK-N-SH细胞中CXCR4表达的变化。结果:FACS检测结果表明CXCR4分子在SK-N-SH肿瘤细胞中高表达,但化疗药物不能降低其在蛋白水平的表达。Real-time PCR结果显示化疗药物也不影响CXCR4在mRNA水平的表达。结论:与我们前期报道化疗药物能影响神经母细胞瘤细胞LA-N-5上CXCR4的表达不同,CTX、THP不能显著降低转移相关趋化因子受体CXCR4在神经母细胞瘤细胞SK-N-SH表面的表达,提示神经母细胞瘤来源不同,其对化疗药物的敏感性不同。OBJECTIVE：To study the expression of CXCR4 on neuroblastoma cells-SK-N-SH and its sensitivity to such chemotherapeutics as Cyclophosphamide（CTX） and Pirarubicin（THP）.METHODS：In vitro culture of neuroblastoma cells-SK-N-SH was conducted with the expression of CXCR4 on SK-N-SH cells detected by flow cytometry.Dose-effects of chemotherapeutics including CTX and THP on SK-N-SH cells were investigated by means of CH50 test.The effects of CTX and THP on CXCR4 expression on SK-N-SH cells at gene and protein levels were detected by Real-time PCR assays and flow cytometry.RESULTS：FACS detection results revealed highly expressed CXCR4 on SK-N-SH cells.Chemotherapeutics CTX and THP were unable to down regulate the expression of CXCR4 on SK-N-SH at protein level.Real-time PCR detection results also showed that CTX and THP had no impact on the expression of CXCR4 at mRNA level.CONCLUSION：In contrary to our previous report in which we claimed that chemotherapeutics can affect（CTX） the expression of chemokine receptor CXCR4 on SK-N-SH cells,in this study we found that CTX and THP couldn＇t significantly lower the expression of chemokine receptor CXCR4 on SK-N-SH cells,suggesting that the sensitivity of neuroblastoma cells to chemotherapy varied with its sources.
